METTL16 exerts an m(6)A-independent function to facilitate translation and tumorigenesis

Rui Su et al.

Summary: METTL16 has dual functions as an m(6)A writer in the nucleus and a translation facilitator in the cytosol, promoting translation through direct interactions with ribosomal RNAs and eukaryotic initiation factors.

RNA demethylase ALKBH5 inhibits TGF-β-induced EMT by regulating TGF-β/SMAD signaling in non-small cell lung cancer

Zelong Sun et al.

Summary: ALKBH5 plays a key role in regulating TGF-beta/SMAD signaling and controlling TGF-beta-induced EMT in NSCLCs. ALKBH5 expression is significantly reduced in metastatic NSCLC and its overexpression inhibits TGF-beta-induced EMT and invasion of NSCLC cells. ALKBH5 affects the expression and mRNA stability of TGF beta R2, SMAD3, and SMAD6 by erasing m(6)A modification in NSCLC cells, and interacts with m(6)A-binding proteins to further regulate TGF-beta signaling.

Multifaceted regulation of translation by the epitranscriptomic modification N6-methyadenosine

Xiao-Min Liu et al.

Summary: m(6)A, an emerging epitranscriptomic code, plays a crucial role in mediating translatomic stability by recruiting translation machinery proteins and serving as a physical barrier, thereby adding an additional layer of complexity to the regulation of translation.

Translation initiation in cancer at a glance

Rachael C. L. Smith et al.

Summary: Cell division, differentiation, and function rely heavily on accurate proteome composition and regulated gene expression. The process of translation, particularly translation initiation, is tightly regulated to control the activity of eIFs and recruit mRNAs to ribosomes. Tumor cells can alter translation initiation mechanisms to promote tumor formation and metastasis.

microRNA-induced translational control of antiviral immunity by the cap-binding protein 4EHP

Xu Zhang et al.

Summary: The mRNA 5' cap-binding protein 4EHP plays a key role in regulating type I IFN production and controlling virus replication. It suppresses IFN-beta production by effecting the miR-34a-induced translational silencing of Ifnb1 mRNA, highlighting an intrinsic regulatory function for miRNA and the translation machinery in maintaining host homeostasis.

Translational repression of NMD targets by GIGYF2 and EIF4E2

Boris Zinshteyn et al.

Summary: Translation of mRNAs with premature termination codons can lead to truncated proteins with deleterious effects, which are prevented by nonsense-mediated mRNA decay (NMD). A genome-wide CRISPR screen identified translational repressors GIGYF2 and EIF4E2 as key regulators of NMD targets, suggesting a model where premature stop codons are recognized and translationally repressed through these factors.

Splice site m6A methylation prevents binding of U2AF35 to inhibit RNA splicing

Mateusz Mendel et al.

Summary: The study reveals that the C. elegans writer METT-10 deposits an m(6)A mark on the SAM synthetase pre-mRNA, inhibiting proper splicing and protein production triggered by a rich diet. Although the mechanism is not present in mammalian SAM synthetase pre-mRNA, splicing inhibition by 3' splice site m(6)A is conserved in mammals.

METTL16 promotes cell proliferation by up-regulating cyclin D1 expression in gastric cancer

Xiao-Kun Wang et al.

Summary: The study showed that METTL16 was highly expressed in gastric cancer and associated with prognosis. In vitro and in vivo experiments confirmed that METTL16 promoted proliferation of GC cells and tumor growth. Furthermore, METTL16 was found to enhance proliferation of GC cells by regulating the expression of cyclin D1.
