The behavior and functions of embryonic microglia

Summary: Microglia, the resident immune cells of the central nervous system, are generated from progenitors in the yolk sac during early embryonic stages and colonize brain structures through migration and proliferation. Despite being a minor population in the developing cortex, microglia interact with neural lineage cells through filopodia extension and migration. Their distribution in the developing brain changes in a stage-dependent manner, playing roles in regulating neurogenesis and neuronal circuit formation.

The presence of arterial vascular smooth muscle cells is crucial for the establishment of perivascular macrophages.

Summary: Green and O'Dea et al. found that early-colonizing microglia in the embryonic zebrafish brain depend on lymphatic vessels for colonization, highlighting the importance of lymphatic cells in microglia development. These early microglia precursors are distinct from the traditionally described microglia and expand preferentially during pathophysiological conditions. The presence of similar microglia in the embryonic brains of mice and humans was also confirmed using single-cell RNA-sequencing datasets.
