Brain-derived autophagosome profiling reveals the engulfment of nucleoid-enriched mitochondrial fragments by basal autophagy in neurons

Juliet Goldsmith et al.

Summary: This study reveals the importance of autophagy in maintaining neuronal health. By isolating autophagic vesicles and using proteomics, the researchers identified mitochondrial proteins as a major cargo engulfed in autophagosomes. They also found that nucleoid-associated proteins are enriched in autophagic engulfment of nucleoid-enriched mitochondrial fragments, and this process requires the mitochondrial fission machinery protein Drp1.

Selective motor activation in organelle transport along axons

Sydney E. Cason et al.

Summary: The active transport of organelles and other cargos along the axon is essential for maintaining neuronal health and function. However, the complex regulatory mechanisms involved in this process are still not fully understood. This review discusses the current understanding of motor regulation in axonal transport and explores how regulatory proteins affect the activity of dynein or kinesin motors. The specific mechanisms that regulate motor activity during organelle trafficking in the axon are also highlighted.

Directing LRRK2 to membranes of the endolysosomal pathway triggers RAB phosphorylation and JIP4 recruitment

Jillian H. Kluss et al.

Summary: Coding mutations in the LRRK2 gene can lead to Parkinson's disease and increase the activity of LRRK2. Lysosomal membrane damage is one mechanism of LRRK2 activation, but it is unclear if activation can occur at other membranes or without membrane damage.

Spatiotemporal analysis of axonal autophagosome-lysosome dynamics reveals limited fusion events and slow maturation

Sydney E. Cason et al.

Summary: In this study, a quantitative model and mathematical modeling were used to investigate the process of autophagosome maturation in neurons. The results showed that fusion between autophagosomes and lysosomes is less frequent during axonal transport, and the breakdown of the inner autophagosomal membrane is slower in neurons, highlighting the importance of this late maturation step.

Parkinson-causing mutations in LRRK2 impair the physiological tetramerization of endogenous α-synuclein in human neurons

Long-term inhibition of mutant LRRK2 hyper-kinase activity reduced mouse brain α-synuclein oligomers without adverse effects

Philip Wing-Lok Ho et al.

Summary: This study demonstrates that long-term inhibition of mutant LRRK2 with GNE-7915 can significantly reduce alpha Syn oligomer accumulation in the striatum without causing adverse peripheral effects. This therapy presents a safe and effective way to ameliorate synucleinopathy in Parkinson's disease.

Mitochondrial dynamics: Shaping and remodeling an organelle network

Adam R. Fenton et al.

Summary: Mitochondria form networks that are continually remodeled and adapted to carry out their cellular function. This remodeling is achieved through changes in morphology, number, and distribution, which are dependent on fission, fusion, shape transition, and transport/tethering along the cytoskeleton. Over the past few years, many mechanisms regulating these processes have been uncovered, showing precise and context-dependent regulation within the cell.

Pathogenic LRRK2 control of primary cilia and Hedgehog signaling in neurons and astrocytes of mouse brain

Shahzad S. Khan et al.

Summary: Activating mutations of LRRK2 cause Parkinson's disease and result in cilia loss. The loss of cilia is associated with phosphorylation of Rab GTPases, defects in astrocytes, and dysregulation of the Hedgehog signaling pathway in the nigrostriatal pathway.

Hyperactive LRRK2 kinase impairs the trafficking of axonal autophagosomes

C. Alexander Boecker et al.

Summary: The hyperactivation of LRRK2 kinase activity is linked to defective axonal autophagosome transport in neurons, particularly in the context of Parkinson's disease. This disruption is caused by the recruitment of SPAG9/JIP4 and subsequent abnormal activation of kinesin-1, leading to impaired autophagosome maturation and degradation. Overall, these findings further establish the significance of defective autophagy in the pathogenesis of PD.

Sequential dynein effectors regulate axonal autophagosome motility in a maturation-dependent pathway

Sydney E. Cason et al.

Summary: The study identifies multiple dynein effectors that regulate the transport and maturation of autophagosomes in neurons, highlighting the tight link between maturation and transport. These findings provide insights into the maintenance of neuronal homeostasis and axonal health through the coordination of dynein-mediated processes.
