Photothermally Triggered Copper Payload Release for Cuproptosis-Promoted Cancer Synergistic Therapy

Summary: In this study, a photothermally triggered nanoplatform (Au@MSN-Cu/PEG/DSF) was constructed to achieve synergistic therapy and overcome the challenges in cancer treatment. The platform released DSF to chelate with Cu2+ and generate highly cytotoxic CuET, inducing cell apoptosis. The Cu+ species formed further promoted toxic mitochondrial protein aggregation, leading to cell cuproptosis. With the combination of photothermal therapy, Au@MSN-Cu/PEG/DSF effectively killed tumor cells and inhibited tumor growth.

The combination of SDT and chemodynamic therapy demonstrated synergistic therapeutic effects both in vitro and in vivo.

Engineering Cu-CuFe2O4 nanoenzyme for hypoxia-relief and GSH-depletion enhanced chemodynamic/sonodynamic therapy

Summary: In this study, a novel Cu-CuFe2O4 nanoenzyme was developed for simultaneous relief of hypoxia and depletion of GSH, enhancing the therapeutic effect of ROS-involved therapy. The nanoenzyme exhibited catalytic activities to continuously generate oxygen from tumor-overexpressed hydrogen peroxide, facilitating hypoxia-relieved sonodynamic therapy. Additionally, the Cu-CuFe2O4 NPs reacted with GSH to generate abundant hydroxyl radicals for chemodynamic therapy. The efficient anticancer ability of Cu-CuFe2O4 NPs was demonstrated in MCF-7 cell killing and multicellular tumor spheroids elimination.

In vivo studies showed a high tumor suppression rate, and the integration of glutamine metabolism therapy with nanocatalytic tumor therapy provides a novel strategy for multimodal tumor treatment.

Copper induces cell death by targeting lipoylated TCA cycle proteins

Summary: Copper is an essential cofactor for all organisms, but excessive copper can lead to cell death. We found that copper-dependent cell death mechanism is different from known mechanisms, and it is related to mitochondrial respiration. Copper directly binds to lipoylated components of the tricarboxylic acid cycle, causing protein aggregation and loss of iron-sulfur cluster proteins, ultimately leading to cell death.

Calcium-chelated nanosystem reversing cancer chemoresistance via replenishing intracellular calcium ions

Summary: Chemoresistance is a major cause of chemotherapy failure and patient death. However, current approaches to reversing chemoresistance are limited. This study developed calcium-chelated nanoparticles that can replenish calcium ions in chemoresistant cancer cells. These nanoparticles released calcium ions more rapidly in the acidic tumor microenvironment, restoring calcium homeostasis and reversing chemoresistance in various cancer cells. Mechanistically, the nanoparticles affected the expression of chemoresistant genes and promoted cancer cell apoptosis through inhibiting the NF-kappa B signaling pathway. These findings provide insight into an effective approach for reversing chemoresistance, suggesting the potential applications of calcium-chelated nanoparticles in treating chemoresistant patients.
