Characterization and structure-property relationships of an injectable thiol-Michael addition hydrogel toward compatibility with glioblastoma therapy

Summary: This study systematically characterized an injectable hydrogel for the treatment of glioblastoma multiforme (GBM), and evaluated the physical and chemical properties as well as biocompatibility of different formulations. The results showed that a hydrogel synthesized with 0.175M NaHCO3 and 50wt% water content exhibited minimal swelling, had a storage modulus suitable for high intracranial pressures, a stable crosslinked polymer network, and was non-toxic to cells with minimal immunogenic responses.

Heterogeneous Glioma Cell Invasion Under Interstitial Flow Depending on Their Differentiation Status

Summary: A deeper mechanistic understanding of heterogeneous glioblastoma cell invasion is crucial for developing therapeutic strategies. Our study revealed that the invasion and protrusion formation of glioma initiating cells (GICs) were significantly enhanced by forward interstitial flow in differentiated cell conditions. The upregulation of integrin beta 1, focal adhesion kinase, and phosphorylated Src was observed, and the flow-induced invasion was significantly inhibited by a Src inhibitor. The flow-induced heterogeneous cell invasion was preceded by nestin-positive GICs at the invasion front and followed by tubulin beta 3-positive differentiated cells.

A novel multidisciplinary approach, based on in vivo experiments in Drosophila and mathematical models, has been proposed to describe the dynamics of active and inactive integrins in relation to matrix metalloprotease concentration and tumor density at the Glioblastoma invasion front, capturing the formation of patterns that drive tumor front evolution.

Cancer Cell-Sticky Hydrogels to Target the Cell Membrane of Invading Glioblastomas

Summary: A cancer cell-sticky hydrogel (CSH) was designed to hinder the invasive motility of glioblastoma cells by modifying the cell membrane with active thiol-enriched interfaces. Cotreatment with the CSH and chemical inhibitors of proinvasive molecules significantly reduced tumor invasion in mouse models, highlighting CSH as a novel therapeutic strategy against glioblastoma.

An alginate-based macroporous hydrogel matrix to trap cancer cells

This model system could be beneficial for drug testing and treatment optimization in glioblastoma.
