Epithelial-to-mesenchymal-like transition events in melanoma

Dennis Pedri et al.

Summary: The review discusses the emergence of a process similar to EMT in melanoma, as well as how comparative analysis with EMT state(s) in epithelial cancers can provide clues for identifying clinically relevant biomarkers for prognosis and new therapeutic strategies.

Pirin, an Nrf2-Regulated Protein, Is Overexpressed in Human Colorectal Tumors

Ying Zhang et al.

Summary: The overexpression of pirin in colorectal cancer is correlated with the activation of Nrf2 and increased expression of NQO1, but not with the expression of AKR1B10 and AKR1C1. Pirin is regulated by Nrf2 and its inhibition can decrease the viability of colorectal cancer cells.

Cancer epigenetics: Past, present and future

Jae Eun Lee et al.

Summary: Over the past 35 years, epigenetic changes have been increasingly recognized as a major driver of carcinogenesis and cancer progression, leading to changes in gene expression that promote the emergence and evolution of cancer cells. Research on the deregulation of chromatin modifiers in cancer cells is shaping our understanding of cancer and guiding the development of novel anticancer therapies targeting epigenetic factors. Areas of particular interest include the actions of emerging epigenetic regulators of carcinogenesis, the tumor microenvironment, and epigenetic tumor heterogeneity.

Role of Pirin, an Oxidative Stress Sensor Protein, in Epithelial Carcinogenesis

Francisco Perez-Dominguez et al.

Summary: Pirin is a protein detected at low levels in normal tissues, but at high levels in multiple cancers, particularly melanomas, cervical cancer, and squamous cell lung carcinomas. Its role in cancer is related to the host response against factors causing oxidative stress, favoring cell migration and metastasis. Pirin is a potential therapeutic target as it is an oxidative stress sensor and transcriptional activator of the NF-kappa B pathway.

Rethinking the biology of metastatic melanoma: a holistic approach

Hendrik H. L. D. Vandyck et al.

Summary: Melanoma-related mortality has remained stable due to treatment failure of metastatic disease and limited knowledge on metastasis formation. Melanoma cells manipulate the tumor microenvironment and express various cell adhesion molecules to promote invasion and metastasis. Studies have shown that primary melanomas shed exosomes to create a favorable premetastatic niche in the microenvironment of secondary organs and lymph nodes.

JARID1-targeted histone H3 demethylase inhibitors exhibit anti-proliferative activity and overcome cisplatin resistance in canine oral melanoma cell lines

Savannah J. Tobin et al.

Summary: Histone demethylases have been found to be overexpressed or have altered activity in human cancers, leading to changes in cell cycle dynamics, DNA repair kinetics, and therapeutic resistance. Targeting histone demethylases has become a significant area of research in human oncology. In a study on canine oral melanoma, it was discovered that histone demethylase inhibitors could reduce cell survival, prolong repair kinetics, and effectively target JARID1B, a highly expressed histone H3 demethylase in canine tumor tissues. These inhibitors also showed anti-proliferative effects in cell lines resistant to platinum-based chemotherapy.
