Peptide backbone-copper ring structure: A molecular insight into copper-induced amyloid toxicity

Summary: Copper ions can promote amyloid diseases associated with amyloid peptides, such as type 2 diabetes, Alzheimer's disease, Parkinson's disease, and amyotrophic lateral sclerosis. By specifically binding to the backbone of T2D-related human islet amyloid polypeptide, Cu2+ can form a ring structure, reduce to Cu+ to produce reactive oxygen species, and modulate amyloid peptide aggregation. This finding sheds new light on the molecular mechanism of Cu2+-induced amyloid toxicity involving enhancement of ROS and modulation of amyloid peptide aggregation.

Summary: Soluble oligomers of the amyloid-beta(1-42) peptide, which are considered neurotoxic in Alzheimer's disease, were characterized using biochemical and biophysical methods. The size of the oligomers was found to correlate with the position of the main amide I' band in the IR spectra, with larger oligomers having lower wavenumbers. Time-resolved aggregation experiments showed a more homogeneous absorption of beta-sheets in the oligomers after several hours, due to the consumption of smaller oligomers.
