Impact of anti-PEG antibody affinity on accelerated blood clearance of pegylated epoetin beta in mice

Tien-Ching Chang et al.

Summary: Antibodies that bind polyethylene glycol (PEG) can accelerate the clearance of pegylated medicines, with higher affinity antibodies leading to greater clearance and reduced drug efficacy. The molar ratio of high affinity anti-PEG antibodies to PEG-EPO correlates with accelerated blood clearance in mice.

Linear Polyglycerol for N-terminal-selective Modification of Interleukin-4

Michael Tully et al.

Summary: Polymer conjugation to biologics is crucial for the development of potent and long acting biotherapeutics. In this study, the potential of linear polyglycerol (LPG) as an alternative polymer to poly(ethylene glycol) (PEG) was investigated. The results showed that LPG and PEG bioconjugates exhibited similar conjugation yields, hydrodynamic sizes, and protein activity. These findings provide promising insights for future pharmacokinetic evaluation of LPG-bioconjugates of therapeutic proteins.

The Effect of Erythropoietin and Its Derivatives on Ischemic Stroke Therapy: A Comprehensive Review

Yuanyuan Ma et al.

Summary: This review provides comprehensive information on the treatment of ischemic stroke with erythropoietin (EPO) and its derivatives. It covers the structure, function, pre-clinical studies, clinical trials, and therapeutic mechanisms of EPO, including its role in suppressing inflammation. The advanced studies on the therapy using EPO derivatives are also discussed. The findings of this review are valuable for basic researchers and clinicians in accelerating the clinical applications of EPO and its derivatives in ischemic stroke.

Polymer selection impacts the pharmaceutical profile of site-specifically conjugated Interferon-?2a

Niklas Hauptstein et al.

Summary: Conjugation of poly(ethylene glycol) (PEG) and linear polyglycerol (LPG) with biologics can improve their pharmacokinetics and efficacy. The study shows that bioconjugates synthesized by specific site PEGylation and LPGylation have better in vitro potency compared to commercial Pegasys, and the overall exposure of 40 kDa PEGylated IFN-α-2a exceeds that of Pegasys.

The evolution of polymer conjugation and drug targeting for the delivery of proteins and bioactive molecules

Antonella Grigoletto et al.

Summary: Polymer conjugation is a leading approach in nanotechnology-based drug delivery systems, aiming to improve in vivo behavior and pharmacokinetic activities of active molecules. While conjugation with biotech drugs has achieved clinical use, conjugation with low molecular weight drugs has not yet reached clinical approval due to challenges in achieving selective targeting. Researchers are working towards identifying more sophisticated and biocompatible delivery approaches to enhance drug targeting for successful clinical applications.
