CRISPR-based VEGF suppression using paired guide RNAs for treatment of choroidal neovascularization

Sook Hyun Chung et al.

Summary: This study compares the effectiveness of using single versus paired gRNAs in the CRISPR-Cas9 system to target the Vegfa gene. Paired gRNAs increased gene ablation rates in human cells but did not enhance VEGF suppression in mouse eyes. Using two gRNAs for genome editing may increase the risk of off-target effects.

Targeted Knockout of the Vegfa Gene in the Retina by Subretinal Injection of RNP Complexes Containing Cas9 Protein and Modified sgRNAs

Andreas Braae Holmgaard et al.

Summary: This study investigates the therapeutic effect of retinal gene therapy using CRISPR/Cas9-mediated genome editing and knockout applications. By utilizing RNPs composed of msgRNAs and SpCas9, efficacy can be enhanced to achieve targeted gene knockout. Moreover, lipoplex delivery of a mixture of RNP and mRNA has been shown to achieve efficient knockout.

Age-related macular degeneration

Monika Fleckenstein et al.

Summary: Age-related macular degeneration (AMD) is the leading cause of legal blindness in the industrialized world, with a complex pathophysiology involving aging, environmental factors, and genetic susceptibility leading to photoreceptor degeneration and drusen formation. Tailoring therapeutics to specific phenotypes and stages may be crucial for preventing irreversible vision loss in the future.
