Fully Symmetric Cyclodextrin Polycarboxylates: How to Determine Reliable Protonation Constants from NMR Titration Data

Summary: The acid-base properties of cyclodextrins persubstituted at C-6 by 3-mercaptopropionic acid were studied using H-1 NMR-pH titrations. Calculation of macroscopic pK(a) values was hindered by severe overlap in protonation steps. By reducing the number of unknown NMR parameters and using the Q-fitting or equidistant macroscopic evaluation approaches, physically unrealistic pK(a) values were obtained. To resolve the issue of cooperativity in carboxylate protonations, the microscopic site-binding model by Borkovec was adapted. However, only one averaged interactivity parameter could be reliably calculated for each CD derivative.

Countermeasures for Preventing and Treating Opioid Overdose

Summary: The only medication available to prevent and treat opioid overdose is naloxone, approved by the FDA nearly 50 years ago. Due to its limitations, a scientific meeting was convened by NIAID/NIH to explore emerging alternative approaches for treating opioid overdose in the event of weaponization of synthetic opioids. This meeting discussed new approaches such as intranasal nalmefene, methocinnamox, covalent naloxone nanoparticles, serotonin receptor agonists, fentanyl-binding cyclodextrin scaffolds, detoxifying biomimetic nanosponge decoy receptors, and antibody-based strategies.

In Vitro and In Vivo Sequestration of Methamphetamine by a Sulfated Acyclic CB[n]-Type Receptor

Summary: Two new acyclic sulfated acyclic CB[n]-type receptors were synthesized and TetM0 was found to have the highest binding affinity towards methamphetamine, fentanyl, MDMA and mephedrone. TetM0 showed no cytotoxicity in vitro and in vivo, and had the potential to reduce hyperactivity in mice treated with methamphetamine.
