Elevated Maternal Folate Status and Changes in Maternal Prolactin, Placental Lactogen and Placental Growth Hormone Following Folic Acid Food Fortification: Evidence from Two Prospective Pregnancy Cohorts

Vitamin B12 and gestational diabetes mellitus: a systematic review and meta-analysis

Summary: This study conducted a systematic review and meta-analysis on the relationship between vitamin B-12 and gestational diabetes mellitus (GDM). The results showed that low vitamin B-12 concentration in the second or third trimester of pregnancy was associated with an increased risk of GDM. Vitamin B-12 deficiency was also found to increase the risk for GDM. Additionally, an increased ratio of high folate to low vitamin B-12 in serum was associated with an increased risk of GDM.

Summary: Folate is an essential dietary micronutrient involved in one-carbon metabolism. The World Health Organisation recommends folic acid (FA) supplementation before conception and during early pregnancy to reduce the risk of fetal neural tube defects. Many countries have implemented mandatory FA fortification policies and recommendations for periconceptional FA supplementation, which have been successful in reducing the incidence of neural tube defects. However, excessive intake of FA has been associated with an increased risk of gestational diabetes mellitus. This article summarizes the current knowledge on the relationship between high FA intake, disrupted one-carbon metabolism, and the pathogenesis of gestational diabetes mellitus, highlighting the need for further research in this area.

Prolactin Is Associated With Insulin Resistance and Beta-Cell Dysfunction in Infertile Women With Polycystic Ovary Syndrome

Multiple linear regression analysis showed that LH, LH/FSH, E-2, FINS, HOMA-IR, and HOMA-beta were negatively associated with serum PRL after adjusting for age and BMI.

Homocysteine inhibits pro-insulin receptor cleavage and causes insulin resistance via protein cysteine-homocysteinylation

Summary: Elevated homocysteine levels are associated with insulin resistance, which is induced by homocysteinylation of the pro-insulin receptor. Homocysteine reacts with and modifies cysteine-825 of pro-IR in the ER, disrupting the original disulfide bond. This modification impairs the interaction between pro-IR and Furin protease in the Golgi apparatus, hindering cleavage of pro-IR and leading to insulin resistance in mice. Inhibiting homocysteinylation could potentially be a new target for drugs to prevent insulin resistance and increase insulin sensitivity.
