Effects of paternal overnutrition and interventions on future generations

Md Mustahsan Billah et al.

Summary: Evidence from human and animal studies suggests that paternal obesity around conception can negatively impact offspring health through developmental programming. This review discusses the current evidence on the impact of paternal obesity on offspring health, focusing on sperm DNA damage as a potential mechanism. Various intervention strategies, including exercise and dietary changes, show promising results in minimizing the effects of paternal obesity on offspring.

Male obesity impacts DNA methylation reprogramming in sperm

Sanaz Keyhan et al.

Summary: Male obesity is associated with altered sperm DNA methylation profiles that appear to affect reprogramming fidelity in a subset of sperm, suggestive of an influence on the spermatogonia. Further work is required to determine the potential heritability of these DNA methylation alterations. If heritable, these changes have the potential to impede normal development.

Neonatal overfeeding during lactation rapidly and permanently misaligns the hepatic circadian rhythm and programmes adult NAFLD

Francesc Ribas-Aulinas et al.

Summary: Childhood obesity is a strong risk factor for adult obesity, type 2 diabetes, and cardiovascular disease. A mouse model of early adiposity was developed through litter size reduction, with mice reared in small litters experiencing obesity, insulin resistance, and hepatic steatosis during adulthood. The study found that the liver played a major role in the development of the disease, with the circadian rhythm being a key factor in the long-term programming of metabolic health.

Genome-wide identification of microRNA targets reveals positive regulation of the Hippo pathway by miR-122 during liver development

Yin Zhang et al.

Summary: Liver development is a complex process involving the interplay of epigenetic regulators, transcription factors, and miRNAs. This study identified the genome-wide miRNA targetomes during mouse liver development and revealed a novel mechanism of terminal differentiation of hepatocytes regulated by the miR-122/Hippo pathway. The findings provide insights into the function of miR-122 in liver pathology, particularly in processes like inflammation, fibrosis, and hepatocellular carcinoma.

Time-Restricted Feeding during Puberty Ameliorates Adiposity and Prevents Hepatic Steatosis in a Mouse Model of Childhood Obesity

Francesc Ribas-Aulinas et al.

Summary: In this study, two short-term chrono-nutritional interventions were explored in a mouse model of childhood obesity, showing that time-restricted feeding is an effective and feasible nutritional intervention to improve metabolic health, especially hepatic steatosis. These findings suggest new potential for safe and efficient chrono-nutritional interventions to enhance metabolic health in children with overweight/obesity.

Paternal multigenerational exposure to an obesogenic diet drives epigenetic predisposition to metabolic diseases in mice

Georges Raad et al.

Summary: Research shows that maintaining a paternal Western diet feeding for five consecutive generations in mice leads to an increase in fat mass and related metabolic diseases over generations, but progenies from these multigenerational Western-diet-fed males develop a healthy overweight phenotype characterized by normal glucose metabolism and without fatty liver, which persists for four subsequent generations. Sperm RNA is suggested to be sufficient for the establishment but not for long-term maintenance of epigenetic inheritance of metabolic pathologies.

Summary: Animal experiments show that high fat diets can harm male reproductive health and increase the risk of chronic diseases in the next generation. The link between dietary habits, sperm epigenetic outcomes, and possible implications for offspring's health are explored in this study.
