Higher stress response and altered quality of life in schizophrenia patients with low membrane levels of docosahexaenoic acid

Vladimir Adrien et al.

Summary: Schizophrenia is a severe mental disorder that affects approximately 1% of the global population. This study aims to identify subgroups of patients with homogeneous symptoms, etiology, prognosis, and treatment response by clustering schizophrenia heterogeneity into various biotypes. The study focuses on a biotype characterized by a specific membrane lipid composition and found that patients with lower levels of omega-3 docosahexaenoic acid (DHA) in their erythrocyte membranes had more hospitalizations, lower quality of life, and higher levels of interleukin-6 and cortisol. These findings highlight the importance of measuring membrane lipid and immunoinflammatory biomarkers in stabilized patients for optimizing interventions and guiding future research.

Potential plasma biomarker panels identification for the diagnosis of first-episode schizophrenia and monitoring antipsychotic monotherapy with the use of metabolomics analyses

Meng Song et al.

Summary: Using liquid chromatography mass spectrometry, the study identified potential biomarkers associated with the diagnosis and treatment of schizophrenia. Biomarker panels were selected to distinguish between schizophrenia and healthy controls, as well as between schizophrenia before and after medication. Disturbances in lipid metabolism, sulfation modification, tryptophan metabolism, anti-inflammatory and antioxidant systems, and unsaturated fatty acids metabolism were observed in schizophrenia. These findings could contribute to the development of objective diagnostic and drug treatment monitoring tools for schizophrenia.

Association between Arachidonic Acid and the Risk of Schizophrenia: A Cross-National Study and Mendelian Randomization Analysis

Yan Gao et al.

Summary: Polyunsaturated fatty acids (PUFAs), especially long-chain PUFAs (LCPUFAs), are essential for cell integrity. This study found an inverse correlation between PUFA consumption and schizophrenia incidence rates, particularly for arachidonic acid (AA) and omega-6 LCPUFAs. Mendelian randomization analyses revealed that genetically predicted AA and gamma-linolenic acid (GLA) were protective factors against schizophrenia. These findings shed new light on the etiology of schizophrenia and suggest a potential dietary supplementation for prevention and treatment.

Reduced erythrocyte membrane polyunsaturated fatty acid levels indicate diminished treatment response in patients with multi-versus first-episode schizophrenia

Summary: This study analyzed the abnormal fatty acid metabolism in multi-episode schizophrenia patients and found that it may contribute to increased treatment resistance to antipsychotic medication. Additionally, certain changes in fatty acid levels/ratios were positively correlated with symptom improvement.

Plasma oxysterols in drug-free patients with schizophrenia

Wassim Guidara et al.

Summary: Evidence suggests a correlation between the profile of oxysterols in plasma and clinical factors in patients with schizophrenia, potentially implicating oxysterols in the pathophysiology of the disorder. The ratio of cholestane-313,5 alpha,613-triol to total cholesterol (Tchol) may be considered an independent risk factor for schizophrenia, while cholestane-313,5 alpha,613-triol, 27-hydroxycholesterol, and cholestanol show promise as potential biomarkers for the disease.

Evidence that complement and coagulation proteins are mediating the clinical response to omega-3 fatty acids: A mass spectrometry-based investigation in subjects at clinical high-risk for psychosis

Subash Raj Susai et al.

Summary: The present study found a long-term association between omega-3 polyunsaturated fatty acids (PUFAs) and changes in complement and coagulation proteins in the blood. Increasing omega-3 PUFAs can alleviate symptom severity and improve cognition in individuals at clinical high risk through modulating effects of complement and coagulation proteins.

EPA and DHA differentially modulate membrane elasticity in the presence of cholesterol

Miranda L. Jacobs et al.

Summary: Polyunsaturated fatty acids (PUFAs) are hypothesized to modulate protein activity by indirectly altering membrane physical properties, with the membrane area expansion modulus (K-a) being a significant controller of channel activity. PUFAs generally decrease the apparent K-a (K-app) of membranes, and in membranes with high levels of cholesterol, docosahexaenoic acid (DHA) has a greater impact on reducing K-app compared to eicosapentaenoic acid (EPA). This study suggests that membrane phase behavior and the distribution of membrane-elasticizing amphiphiles affect the ability of a membrane to stretch.

Shared Biological Pathways between Antipsychotics and Omega-3 Fatty Acids: A Key Feature for Schizophrenia Preventive Treatment?

Ariel Frajerman et al.

Summary: Schizophrenia typically emerges during adolescence and the detailed pathophysiology and treatment of the disease remain relatively unknown. Antipsychotics are effective for first episode psychosis and chronic schizophrenia, but not for ultra-high risk patients. Omega-3 supplementation has shown potential benefits in preventing and treating schizophrenia in early stages by modulating various biological systems.

Probability of Transition to Psychosis in Individuals at Clinical High Risk An Updated Meta-analysis

Summary: This study quantitatively analyzed the risk of transitioning from CHR-P individuals to psychosis, finding that the transition risk continues to increase in both short and long terms. Lower proportion of female individuals and higher frequency of brief psychotic symptoms were associated with increased transition risk.

Associations between plasma fatty acid concentrations and schizophrenia: a two-sample Mendelian randomisation study

Hannah J. Jones et al.

Summary: The study indicated that long-chain omega-3 and omega-6 fatty acid concentrations were associated with a lower risk of schizophrenia, while short-chain omega-3 and omega-6 fatty acids were weakly linked to an increased risk of schizophrenia. Further research is needed to investigate whether supplementation of long-chain polyunsaturated fatty acids may help prevent schizophrenia onset.
