Bi-directional Control of Synaptic Input Summation and Spike Generation by GABAergic Inputs at the Axon Initial Segment

Ziwei Shang et al.

Summary: Different from other types of inhibitory interneurons, chandelier or axo-axonic cells only form synapses onto the axon initial segment (AIS) of targeted pyramidal cells. The timing relationship between AIS-GABAergic inputs and dendritic excitatory glutamatergic inputs determines their regulation of synaptic integration and spike generation. When AIS-GABAergic inputs precede glutamatergic inputs by more than 15 ms, they enhance the excitatory inputs, while for nearly simultaneous excitatory inputs, they primarily produce shunting inhibition.

Regulation of Axon Initial Segment Diameter by COUP-TFI Fine-tunes Action Potential Generation

Xuanyuan Wu et al.

Summary: COUP-TFI is a crucial regulator of AIS diameter in both developing and adult mouse neocortex, influencing changes in AIS diameter and impairment of AP generation. Overexpression of COUP-TFI increases AIS diameter and facilitates AP generation, but weakens the receiving spontaneous network.

Glucose increases the length and spacing of the lattice structure of the axon initial segment

Yiming Wang et al.

Summary: Glucose triggers the plasticity of axon initial segment (AIS) in cultured neurons. Structured illumination microscopy imaging reveals an increased spacing in the AIS lattice structure under glucose treatment. The 480-kDa AnkG plays a crucial role in AIS plasticity.

Single-neuron projectome of mouse prefrontal cortex

Le Gao et al.

Summary: This study reveals the organization principles of axon projections within and outside the prefrontal cortex (PFC) using single-neuron reconstruction. It identifies different subtypes of PFC projection neurons and uncovers topographic rules and target selection patterns. The study also provides a hierarchical map within PFC and identifies three functionally related PFC modules. The findings highlight the importance of neuronal connectivity in understanding diverse PFC functions.

Differential encoding in prefrontal cortex projection neuron classes across cognitive tasks

Jan H. Lui et al.

Summary: Single-cell transcriptomics has been used to classify neurons in the mammalian brain, and in this study, researchers examined how specific transcriptomic types of mouse prefrontal cortex (PFC) projection neurons are related to axonal projections and encoding properties across multiple cognitive tasks. They found that most types of neurons projected to multiple targets, and that task signals are organized redundantly but with clear quantitative biases across cells of specific molecular-anatomical characteristics.

Brief Sensory Deprivation Triggers Cell Type-Specific Structural and Functional Plasticity in Olfactory Bulb Neurons

Elisa Galliano et al.

Summary: Temporary sensory deprivation can induce structural and functional plasticity in dopaminergic neurons in the olfactory bulb, resulting in changes in axon initial segment length and intrinsic excitability. This experience-dependent plasticity may help fine-tune sensory processing in the face of continuously fluctuating inputs.

Postnatal connectomic development of inhibition in mouse barrel cortex

Anjali Gour et al.

Summary: The study quantifies the circuit patterns of synaptogenesis for inhibitory interneurons in the developing mouse somatosensory cortex, revealing different developmental profiles and time courses for synaptic innervation of cell bodies, apical dendrites, and axon initial segments. Apical dendrites are innervated early and specifically, while axons innervating cell bodies gradually acquire specificity over time.

Sensory input drives rapid homeostatic scaling of the axon initial segment in mouse barrel cortex

Nora Jamann et al.

Summary: The study reveals bidirectional AIS plasticity in cortical neurons, demonstrating that the AIS acts in vivo as a homeostatic regulatory domain in a behaviorally relevant context, with structural changes in the AIS length influencing neuronal excitability and action potential generation.

Structure and function of axo-axonic inhibition

Casey M. Schneider-Mizell et al.

Summary: This study used electron microscopy and functional imaging to investigate the connectivity of chandelier cells in the mouse visual cortex, finding that synapse number is highly variable across the population and correlated with structural features of target neurons. Biophysical simulations demonstrated the effectiveness of axo-axonic inhibition in controlling excitatory output, especially when excitation and inhibition are co-active. In vivo chandelier cell activity was found to be highly correlated with pupil dilation, suggesting that chandelier cells provide a circuit-wide signal adjusted relative to target neuron properties.

Diversity amongst human cortical pyramidal neurons revealed via their sag currents and frequency preferences

Homeira Moradi Chameh et al.

Summary: The study reveals the diverse electrophysiological properties of human cortical pyramidal neurons, with L5 pyramidal cells being the most excitable and exhibiting the most prominent sag current. While subthreshold resonance is more common in L3c and L5, L5 neurons are uniquely adept at tracking both delta and theta frequency inputs. These biophysical properties may play a crucial role in the generation of cortical theta oscillations.
