3D strain analysis of trabecular bone within the osteoarthritic humeral head subjected to stepwise compressive loads

Jonathan Kusins et al.

Summary: The study utilized DVC technique to investigate the local load carrying characteristics of trabecular bone in OA patients' humeral heads, finding significantly higher third principal strains in regions that eventually fractured. Bone volume fraction, trabecular separation, and trabecular number were found to influence the likelihood of trabecular bone fracture.

A 3D in Silico Multi-Tissue Evolution Model Highlights the Relevance of Local Strain Accumulation in Bone Fracture Remodeling

Camille Perier-Metz et al.

Summary: A computer model based on mechanical strain accumulation was proposed to investigate the mechanobiological regulation of bone remodeling during the late phase of healing. The model successfully predicted the reopening of the bone marrow cavity and the resorption of the external callus. The results suggest that local strain accumulation can explain the fracture remodeling process and this mechanobiological response is conserved among different mammal species.

Characterizing the Mechanical Behavior of Bone and Bone Surrogates in Compression Using pQCT

Johannes D. Pallua et al.

Summary: The study analyzed the process of structural failure in bones using micro-compression and micro-computed tomographic imaging. It found that by creating 3D models and calculating various parameters, it is possible to precisely analyze the areas of trabecular failure and the load conditions.

Clinical observation of diminished bone quality and quantity through longitudinal HR-pQCT-derived remodeling and mechanoregulation

Caitlyn J. Collins et al.

Summary: High resolution peripheral quantitative computed tomography (HR-pQCT) combined with longitudinal imaging protocol and finite element analysis can aid in the early diagnosis of bone disease, and assess bone formation, resorption, and the relationship with mechanical loading. This study found that HR-pQCT can detect subtle changes in bone quantity and quality that traditional measures cannot capture.

Biophysical Modulation of Mesenchymal Stem Cell Differentiation in the Context of Skeletal Repair

Clark T. Hung et al.

Summary: The skeleton has the ability to remodel and repair itself in response to biophysical stimuli, allowing it to adapt to its physiological roles. Cells in the skeleton sense and respond to these stimuli to meet the demands of the organism. Mechanical strain is the most recognized stimulus, but other signals such as fluid flow and ion movements also play a role. The application of electrical and electromagnetic fields can be used to study the effects of these stimuli on cell differentiation and extracellular matrix formation.

Scaffold Pore Geometry Guides Gene Regulation and Bone-like Tissue Formation in Dynamic Cultures

Marina Rubert et al.

Summary: The study demonstrates that cell activity and bone-like tissue formation are dependent on scaffold pore geometry and mechanical environment. Dynamic culture conditions promote regular extracellular matrix formation and mineral distribution. Scaffold pore geometry plays a crucial role in osteoblast cell differentiation and bone-like tissue formation.

Alterations in osteocyte lacunar morphology affect local bone tissue strains

Haniyeh Hemmatian et al.

Summary: This study found that changes in lacunar volume and orientation can increase strains in osteocytes and bone tissue, while increased lacuna sphericity leads to decreased effective strains. Additionally, old mice with smaller and more round lacunae experience lower local bone tissue strains compared to young mice. Understanding the impact of alterations in lacunar morphology on osteocytes is important for investigating impaired bone mechanoresponsiveness with aging.

Microstructure Determines Apparent-Level Mechanics Despite Tissue-Level Anisotropy and Heterogeneity of Individual Plates and Rods in Normal Human Trabecular Bone

Y. Eric Yu et al.

Summary: This study investigated the tissue mineral density (TMD)-dependent elastic modulus of individual trabeculae and found significantly higher TMD and tissue modulus in trabecular plates than in rods. Axial tissue modulus was consistently higher than lateral tissue modulus regardless of trabecular type and orientation, with axial correlation predicting higher tissue modulus than lateral correlation at the same TMD level.

Biophysical Stimuli as the Fourth Pillar of Bone Tissue Engineering

Zhuowen Hao et al.

Summary: The repair of critical bone defects remains a challenge globally. Three canonical pillars of bone tissue engineering have been widely used but have drawbacks, while biophysical stimuli show potential as the fourth pillar. Different types of biophysical stimuli can induce osteogenesis of mesenchymal stem cells and direct their osteogenic differentiation through biophysical transduction mechanisms. Different application strategies of biophysical stimuli in bone tissue engineering are also presented, along with discussions on ongoing challenges and future perspectives.
