The Obscure Potential of AHNAK2

Mohamed Zardab et al.

Summary: AHNAK2 is a newly discovered protein with a strong association with oncogenesis in various epithelial cancers. It has a tripartite structure and is believed to be involved in the formation of large multi-protein complexes. High expression of AHNAK2 is found in several cancers, including clear cell renal carcinoma and pancreatic ductal adenocarcinoma, and is associated with poor prognosis. Limited research has been conducted on the function and relation of AHNAK2 with cancer, but early studies suggest its potential as a future biomarker and therapeutic target.

The N6-methyladenosine modification enhances ferroptosis resistance through inhibiting SLC7A11 mRNA deadenylation in hepatoblastoma

Li Liu et al.

Summary: In this study, the researchers found that METTL3-mediated SLC7A11 m6A modification enhanced resistance to ferroptosis in hepatoblastoma (HB). Mechanistically, SLC7A11 mRNA exhibited abnormal METTL3-mediated m6A modification, which enhanced its stability and expression. IGF2 mRNA-binding protein 1 (IGF2BP1) was identified as the m6A reader of SLC7A11, enhancing SLC7A11 mRNA stability and expression by inhibiting SLC7A11 mRNA deadenylation in an m6A-dependent manner. The study highlights the critical role of the m6A modification in SLC7A11-mediated ferroptosis and provides a potential strategy for HB therapy through blockade of the m6A-SLC7A11 axis.

Reduced IQGAP2 Promotes Bladder Cancer through Regulation of MAPK/ERK Pathway and Cytokines

Fei Song et al.

Summary: The study revealed that downregulation of IQGAP2 in bladder cancer can inhibit cell growth by suppressing proliferation, while reduced IQGAP2 promotes cancer cell migration and may affect growth and metastasis through cytokine regulation. This suggests a tumor-suppressing role of IQGAP2 in bladder cancer, potentially through inhibiting the MAPK/ERK pathway and reducing cytokines.

Mutation landscape ofTSC1/TSC2in Chinese patients with tuberous sclerosis complex

Yuhuan Meng et al.

Summary: Genetic testing of TSC1 and TSC2 is crucial for diagnosing tuberous sclerosis complex (TSC), revealing a comprehensive mutation landscape in this study. The research also identified potential risk non-TSCs variants present in patients with no mutations detected in TSC1/TSC2.

Methyltransferase-like 1 regulates lung adenocarcinoma A549 cell proliferation and autophagy via the AKT/mTORC1 signaling pathway

Chen Wang et al.

Summary: The study reveals that METTL1 is upregulated in LUAD tissues and associated with unfavorable prognosis, promoting proliferation and colony formation of A549 cells while inhibiting autophagy. It suggests that METTL1 acts as an oncogene in LUAD and may be a potential prognostic predictor and therapeutic target.

Reduced IQGAP2 expression promotes EMT and inhibits apoptosis by modulating the MEK-ERK and p38 signaling in breast cancer irrespective of ER status

Dinesh Kumar et al.

Summary: IQGAP2 functions as a tumor suppressor in breast cancer by enhancing apoptosis, reducing epithelial-mesenchymal transition, and negatively regulating oncogenic properties of breast cancer cells. The interaction between IQGAP1 and IQGAP2 suggests the possibility of IQGAP2 sequestering IQGAP1 to inhibit ERK activation. Targeting IQGAP2 could be a potential therapeutic intervention for breast cancer.
