Design and Synthesis of New Quinoxaline Derivatives as Potential Histone Deacetylase Inhibitors Targeting Hepatocellular Carcinoma: In Silico, In Vitro, and SAR Studies

Chao Ma et al.

Summary: Guided by the structural optimization principle and the promising anticancer effect of the quinoxaline nucleus, a new series of novel HDAC inhibitors were designed and synthesized. The synthesized compounds showed promising anti-proliferative activities against liver cancer cell lines and were further evaluated for their effects on gene expression levels and HDAC inhibition. Compound 6(c) emerged as the most potent candidate, demonstrating significant cytotoxic activities and inhibitory effects on HDAC1, HDAC4, and HDAC6, with characteristics warranting further study for future drug development.
