Carboxyl Methyltransferase Catalysed Formation of Mono- and Dimethyl Esters under Aqueous Conditions: Application in Cascade Biocatalysis

Lucy C. Ward et al.

Summary: Carboxyl methyltransferase enzymes from Aspergillus fumigatus showed high regioselectivity and good conversions in methylating a broad range of aromatic mono- and dicarboxylic acids. The second methylation of dicarboxylic acids exhibited strong pH dependence, with an optimum at pH 5.5-6. Potential for industrial biotechnology application was demonstrated in the production of a bioplastics precursor from bioderived 5-hydroxymethylfurfural.

Rational hinge engineering of carboxylic acid reductase from Mycobacterium smegmatis enhances its catalytic efficiency in biocatalysis

Liuzhu Wang et al.

Summary: A carboxylic acid reductase (CAR) originating from Mycobacterium smegmatis was redesigned through rational hinge engineering to improve catalytic efficiency. Mutagenesis of the hinge region resulted in a mutant showing a 6.57-fold increase in catalytic efficiency. Molecular dynamics simulations revealed the key interactions leading to enhanced activity were due to strong binding of the acyl-AMP complex. This two-step strategy provided useful references for the engineering of CARs and other similar enzymes with multiple domains.

Synthesis and Hemostatic Activity of New Amide Derivatives

Lukasz Banach et al.

Summary: Eight dipeptides containing antifibrinolytic agents were successfully synthesized using a three-step process. Hemolysis tests and clotting tests showed that these compounds had no significant effects on blood components and clotting time. The clot formation and fibrinolysis assay revealed that two of the compounds exhibited high activity in the plasma coagulation cascade and fibrinolytic efficiency. The cytotoxicity and genotoxicity evaluation demonstrated that the synthesized dipeptides caused minimal damage to cells and DNA.
