The most common linkers in bioactive molecules and their bioisosteric replacement network

Redox Activity of Iron Diazine-Diimine Carbonyl and Dinitrogen Complexes: A Comparative Study of the Influence of the Heterocyclic Ring

Summary: A detailed investigation of the electronic structure of diazinediimine iron complexes and their comparison with pyridine analogues reveals subtle but important differences, including the impact of the supporting heterocycle on redox potentials. Different electron-transfer processes are affected by the differences in pi-acidity of the supporting heterocycle, highlighting the importance of the supporting ligand in the electronic properties of the complexes. The reduction of LFe(CO)(2) can lead to either ligand-centered radicals or C-C-bonded dimers, with observable reversible cleavage of the C-C bond in the latter case, showcasing the versatility of these complexes in redox chemistry.

Aqueous Amine-Tolerant [2+2] Photocycloadditions of Unactivated Olefins

Summary: The Kochi-Salomon reaction is a photochemical [2+2] cycloaddition that can combine two electronically unactivated olefins into a cyclobutane. However, this reaction remains largely unexplored and is known for its intolerance to Lewis/Bronsted basic amines and amides. In this study, we disclose a transformation that can be carried out in water using readily available Cu(II) salts and mineral acids. Additionally, we demonstrate the application of this methodology in synthesizing a variety of amine-containing cyclobutanes, including known and novel pharmacological analogues.

Bicyclic Piperidines via [2+2] Photocycloaddition

Summary: A synthetic strategy for fused bicyclic piperidines, used as building blocks in medicinal chemistry, was developed. The key step involved an intramolecular [2 + 2]-photocyclization, performed on a gram scale. Crystallographic analysis of the obtained compounds revealed that they occupy a novel chemical space and can be considered as elongated analogues of 3-substituted piperidines.

Iron-Catalyzed Trimerization of Terminal Alkynes Enabled by Pyrimidinediimine Ligands: A Regioselective Method for the Synthesis of 1,3,5-Substituted Arenes

Summary: The development of pyrimidine-based analogues of PDI iron complexes has enabled a functional-group-tolerant methodology for the catalytic trimerization of terminal aliphatic alkynes, leading to 1,3,5-substituted arenes as the main products. The enhanced pi-acidity of the pyrimidine ring, combined with the hemilability of the imine groups coordinated to the iron center, plays a key role in facilitating this transformation. The catalytic reaction proceeds via a 1,3-substituted metallacycle, explaining the observed 1,3,5-regioselectivity.
