Presynaptic targeting of botulinum neurotoxin type A requires a tripartite PSG-Syt1-SV2 plasma membrane nanocluster for synaptic vesicle entry

Summary: It has been found that botulinum neurotoxin type A (BoNT/A) targets nerve terminals by binding to two receptors on the neuronal plasma membrane: polysialoganglioside (PSG) and synaptic vesicle glycoprotein 2 (SV2). The entry of BoNT/A into synaptic vesicles (SVs) requires a tripartite surface nanocluster, which consists of a preassembled PSG-synaptotagmin-1 (Syt1) complex and SV2 on the neuronal plasma membrane. This tripartite nanocluster controls the endocytic sorting of the toxin into synaptic vesicles.

The P301L mutant Tau promotes abnormal clustering of Fyn in dendrites, and inhibition of Fyn's auto-inhibitory closed conformation increases its clustering. The ability of P301L Tau to form biomolecular condensates enhances Fyn clustering and signaling.

A framework for evaluating the performance of SMLM cluster analysis algorithms

Cluster analysis is an effective method for extracting meaningful information from single-molecule localization microscopy data, but there is no consensus framework for evaluating the performance of different algorithms. This study proposes a systematic approach and evaluation metrics based on simulated conditions to score the success of clustering algorithms.

Three distinct populations of IL-2R gamma clusters were identified, with different sizes playing different roles in IL-2R internalization and signaling. Membrane cholesterol and the branched actin cytoskeleton were found to be key regulators of IL-2R gamma clustering and IL-2-induced signaling, affecting CIE initiation and signal transduction.
