Summary: A small molecule tubulin probe has been developed for use in single-molecule localization microscopy, STED microscopy, and MINFLUX nanoscopy, enabling measurement of microtubule diameter in living and fixed cells. The probe, HMSiR-tubulin, allows for complementary use of different nanoscopy techniques and has demonstrated utility in resolving inner microtubule diameter for the first time.

Modular Synthetic Approach to Silicon-Rhodamine Homologues and Analogues via Bis-aryllanthanum Reagents

Summary: This paper presents a modular synthetic approach for preparing analogues of the far-red fluorophore silicon-rhodamine (SiR), using a regioselective double nucleophilic addition of aryllanthanum reagents to esters, anhydrides, and lactones to improve functional group tolerance. It represents a unified strategy for preparing cell-permeant SiR fluorescent labels. In combination with Pd-catalyzed hydroxy- or aminocarbonylation, it provides a streamlined synthetic pathway for a series of validated live-cell-compatible fluorescent dyes.

Kinetic and Structural Characterization of the Self-Labeling Protein Tags HaloTag7, SNAP-tag, and CLIP-tag

Summary: The study compared the labeling kinetics and substrate specificities of self-labeling protein tags (SLPs) HaloTag7, SNAP-tag, and CLIP-tag. It was found that HaloTag7 reaches almost diffusion-limited labeling rate constants with some substrates, while SNAP-tag labeling rate constants are less affected by the structure of the label. Crystal structures of HaloTag7 and SNAP-tag labeled with fluorescent substrates helped explain their substrate preferences and aided in the design of substrates with improved labeling kinetics.
