Emerging roles of ferroptosis in infectious diseases

Salimata Bagayoko et al.

Summary: Lipid peroxidation is a continuously occurring cellular process in living organisms, affecting various physiological and pathological contexts. Polyunsaturated fatty acids are a major target of oxidation, especially in membranous phospholipids. Recent studies have identified ferroptosis as an iron- and lipid peroxidation-dependent cell necrosis, with implications in a wide range of physiological and pathological processes, such as cancer, neurodegenerative diseases, and metabolic disorders.

M. tuberculosis PknG manipulates host autophagy flux to promote pathogen intracellular survival

Pupu Ge et al.

Summary: PknG promotes macroautophagy/autophagy induction by competitively binding to AKT and inhibits autophagosome maturation through targeting RAB14, resulting in blocked autophagy flux and enhanced pathogen intracellular survival.

Ferroptosis: machinery and regulation

Xin Chen et al.

Summary: Ferroptosis is an iron-dependent, non-apoptotic form of regulated cell death caused by lipid peroxidation. Lipoxygenase is the main promoter of ferroptosis, while GPX4 is the central repressor of this process.

PRMT6 methylation of RCC1 regulates mitosis, tumorigenicity, and radiation response of glioblastoma stem cells

Tianzhi Huang et al.

Summary: This study reveals that a CK2 alpha-PRMT6-RCC1 signaling axis plays a crucial role in the stem-like properties and tumor biology of GSCs, contributing to chromatin condensation, mitosis, and overall malignancy in GBM. EPZ020411, a specific small-molecule inhibitor for PRMT6, shows potential as a therapeutic target for treating GBM by suppressing RCC1 arginine methylation and enhancing the efficacy of radiotherapy against GSC brain tumor xenografts.

Multifaceted mechanisms mediating cystine starvation-induced ferroptosis

Zhennan Shi et al.

Summary: Recent studies have shown additional mechanisms behind cystine starvation-induced ferroptosis, apart from impaired GSH synthesis, highlighting the importance of the cyst(e)ine/glutathione/glutathione peroxidase 4 (GPX4) axis in triggering ferroptosis and inhibiting tumor growth.
