ArpC5 isoforms regulate Arp2/3 complex-dependent protrusion through differential Ena/VASP positioning

Florian Fassler et al.

Summary: Regulation of the Arp2/3 complex plays a crucial role in the nucleation of branched actin networks. Incorporation of subunit isoforms, such as ArpC5 and ArpC5L, fine-tunes nucleation activity and branch junction stability, affecting cell migration. Furthermore, ArpC5 isoforms impact the positioning of Ena/VASP proteins, which mediate isoform-specific effects on actin assembly and contribute to a signaling pathway enhancing cell migration.

IL-6 regulates induction of C-reactive protein gene expression by activating STAT3 isoforms*

Donald N. Ngwa et al.

Summary: C-reactive protein (CRP) is synthesized in hepatocytes and its serum concentration increases during the acute phase response. The combination of IL-6 and IL-10 leads to maximal CRP expression in human hepatoma Hep3B cells. The transcription factor STAT3, particularly its alpha isoform, plays a crucial role in inducing CRP expression by binding to the consensus STAT3-binding sites on the CRP promoter. IL-6 activates multiple isoforms of STAT3, including STAT3 alpha and STAT30, which regulate the induction of CRP expression through all four STAT3 sites.

ARPC1B binds WASP to control actin polymerization and curtail tonic signaling in B cells

Gabriella Leung et al.

Summary: The deficiency of ARPC1B can disrupt the nucleation of ARP2/3 complexes by WASP, affecting the cytoskeletal structures in B cells and macrophages, leading to increased tonic signaling in immune cells and impacting the threshold for activation in response to microbial-associated molecular patterns. This highlights the critical role of ARPC1B in controlling steady-state signaling of immune cells.

Therapeutic targeting of IL-6 trans-signaling

Stefan Rose-John

Summary: IL-6 is a cytokine involved in immunity, neural cell maintenance, and metabolism. It is synthesized by various cells and its levels are elevated during infection and inflammation. IL-6 binds to a receptor complex and can also act on cells without the IL-6 receptor through trans-signaling.
