5-hydroxytryptamine produced by enteric serotonergic neurons initiates colorectal cancer stem cell self-renewal and tumorigenesis

Summary: This study reveals the importance of intestinal nerve cells in the self-renewal of colorectal cancer stem cells (CSCs) and the development of colorectal tumors. Serotonergic neurons in the enteric system produce serotonin (5-HT) as a regulator of CSC self-renewal. 5-HT interacts with HTR1B/1D/1F receptors to activate Wnt/β-catenin signaling in colorectal CSCs. Additionally, a metabolite derived from the colorectal cancer-enriched microbiota, isovalerate, suppresses the enrichment of the NuRD complex on the Tph2 promoter, leading to the expression of Tph2 and subsequent 5-HT production. 5-HT signaling is correlated with the severity of colorectal cancer.

rtcisE2F promotes the self-renewal and metastasis of liver tumor-initiating cells via N6-methyladenosine-dependent E2F3/E2F6 mRNA stability

Summary: Liver cancer is a highly heterogeneous disease with various cell types present in tumor tissue. Liver tumor initiating cells (TICs) play significant roles in tumor heterogeneity, initiation, and metastasis. A recently discovered functional circular RNA called rtcisE2F is found to be highly expressed in liver cancer and liver TICs. rtcisE2F is involved in the self-renewal and activities of liver TICs by targeting E2F6 and E2F3 mRNA and regulating their expression. Mechanistically, rtcisE2F acts as a scaffold for the interaction between N-methyladenosine (m(6)A) reader IGF2BP2 and E2F6/E2F3 mRNA, affecting their stability and decay. Additionally, E2F6 and E2F3 are found to be essential for liver TIC self-renewal and Wnt/beta-catenin activation.

circREEP3 Drives Colorectal Cancer Progression via Activation of FKBP10 Transcription and Restriction of Antitumor Immunity

Summary: Colorectal cancer (CRC) is a common tumor worldwide, and circular RNA circREEP3 is found to be upregulated in CRC tissues. Upregulation of circREEP3 predicts poor patient survival. Mechanistically, circREEP3 activates FKBP10 transcription to promote CRC tumorigenesis and metastasis, while restricting RIG-1-dependent antitumor immunity.

This mechanistic link between aberrant cell cycle and glycolysis addiction provides insights into the coupling of metabolic fluctuation with periodic cell cycle in mammalian cells.
