Aptamers Entirely Built from Therapeutic Nucleoside Analogues for Targeted Cancer Therapy

Lijuan Zhu et al.

Summary: Aptamer-drug conjugates (ApDCs) are a promising drug delivery system for targeted cancer therapy. However, the conventional ApDC has limited drug loading capacity. To address this challenge, researchers have developed drugtamers, whole drug-constituted DNA-like oligomers, which have 100% drug loading ratio, active targeting capability, and enzyme-mediated release of active therapeutics.

Phosphorothioate nucleic acids for probing metal binding, biosensing and nanotechnology

Runjhun Saran et al.

Summary: Phosphorothioate modification, which replaces one of the non-bridging oxygen atoms by sulfur in the phosphate backbone of nucleic acids, has various applications in the fields of nucleic acids research, surface science, and metal binding probes. The ability to graft functional groups onto the sulfur atom and the presence of chiral phosphorus centers at each modification site highlight the versatility and potential of phosphorothioate nucleic acids in various research areas.

Functional Amphiphilic Poly(2-oxazoline) Block Copolymers as Drug Carriers: the Relationship between Structure and Drug Loading Capacity

Si Dong et al.

Summary: Poly(2-oxazoline) (POx) is a type of polymeric amide with promising cyto- and hemo-compatibility, and it can be used as drug carriers. The drug loading capacity (DLC) of POx can be improved by modifying functional groups, and a series of functional POxs provide options for various drug loadings. The encapsulation of drugs with different characteristics can be achieved by selecting suitable functional POxs.

Delivery of local anaesthetics by a self-assembled supramolecular system mimicking their interactions with a sodium channel

Tianjiao Ji et al.

Summary: A self-assembling delivery system was developed for site-1 sodium channel blockers to prolong nerve blockade and reduce systemic toxicity. The system mimicked specific interactions of the blockers with sodium channel peptides, utilizing modified peptides with hydrophobic domains for nanofiber assembly. The strategy could potentially be applied to designing drug delivery systems for receptor-mediated drugs.
