Hypoxia signaling in cancer: Implications for therapeutic interventions

Yan Zhuang et al.

Summary: Hypoxia is a persistent physiological feature of solid tumors, driving malignancy and reducing the effectiveness of other tumor therapies. The critical role of hypoxia in tumor development, including metabolism, immunity, and angiogenesis, is discussed. Treatment methods for hypoxic tumor microenvironment, including hypoxia-targeted therapy and improving oxygenation, are summarized in this review.

Harnessing cytokines and chemokines for cancer therapy

David J. Propper et al.

Summary: Cytokines play a crucial role in cancer development and progression. While cytokines have been studied for their therapeutic potential in oncology, clinical trials have shown limited efficacy. However, the advent of immunotherapies and a better understanding of the tumor microenvironment have opened up new possibilities for harnessing cytokine networks in cancer treatment.

COAST: An Open-Label, Phase II, Multidrug Platform Study of Durvalumab Alone or in Combination With Oleclumab or Monalizumab in Patients With Unresectable, Stage III Non-Small-Cell Lung Cancer

Roy S. Herbst et al.

Summary: The COAST study evaluated the efficacy of durvalumab combined with other antibodies in patients with unresectable stage III non-small-cell lung cancer. The results showed that compared to durvalumab alone, the combination of durvalumab with oleclumab or monalizumab significantly improved the objective response rate and progression-free survival of patients.

CCR2/CCR5 inhibitor permits the radiation-induced effector T cell infiltration in pancreatic adenocarcinoma

Jianxin Wang et al.

Summary: This preclinical study demonstrates the potential of CCR2/CCR5 dual-antagonist in counteracting radiation-induced suppressive signals and enhancing the effector T cell pathway. It supports the ongoing clinical trial of combining this therapy with radiation and anti-PD-1 antibody for locally advanced pancreatic cancer.

Regulation of gene expression by glycolytic and gluconeogenic enzymes

Xueli Bian et al.

Summary: Gene transcription and cell metabolism are two interrelated biological processes, with altered glucose metabolic gene expression being a major driver of enhanced aerobic glycolysis in tumor cells. Notably, glycolytic and gluconeogenic enzymes in tumor cells also have additional functions and directly regulate gene expression by modulating chromatin or transcriptional complexes. The mutual regulation between cellular metabolism and gene expression is a unique feature of tumor cells and provides specific molecular and functional targets for cancer treatment.

Oncolytic peptide LTX-315 induces anti-pancreatic cancer immunity by targeting the ATP11B-PD-L1 axis

Tianyu Tang et al.

Summary: This study demonstrates that LTX-315 can enhance lymphocyte infiltration and induce an antitumor immune response by inhibiting PD-L1 expression in pancreatic tumors. ATP11B is identified as a potential target of LTX-315 and a critical regulator in maintaining PD-L1 expression.

TGFβ biology in cancer progression and immunotherapy

Rik Derynck et al.

Summary: TGF beta signaling plays a key role in cancer progression by promoting cancer cell invasion, dissemination, stem cell properties, therapeutic resistance, and the generation of an immunosuppressive environment. Inhibiting TGF beta signaling is considered a crucial avenue to enhance the efficacy of cancer immunotherapies. Understanding and targeting TGF beta signaling mechanisms in tumors and their microenvironment may provide valuable insights for cancer treatment.

NAD+ Metabolism Maintains Inducible PD-L1 Expression to Drive Tumor Immune Evasion

Hongwei Lv et al.

Summary: The study reveals that NAMPT enzyme drives PD-L1 expression in various tumors and governs tumor immune evasion through CD8(+) T cell regulation. NAD(+) metabolism maintains the activity and expression of Tet1, which plays a critical role in PD-L1 expression on tumors.
