Phase separation properties of RPA combine high-affinity ssDNA binding with dynamic condensate functions at telomeres

Vincent Spegg et al.

Summary: The replication protein A (RPA) tends to assemble into dynamic condensates, which are stimulated by sub-stoichiometric amounts of single-stranded DNA (ssDNA) and regulated by RPA2 phosphorylation. These RPA condensates play a functional role in telomere clustering and RAD52-dependent telomere maintenance.

Cryo-EM structure of the human CST-Polα/primase complex in a recruitment state

Sarah W. Cai et al.

Summary: This study reports a cryo-EM structure of human CST-Pol alpha/primase and reveals an evolutionarily conserved interaction between POLA1 and CTC1. The binding mode of CST to the N-terminus of POLA1 is revealed by negative-stain EM and cross-linking mass spectrometry. In addition, disease mutations associated with Coats plus syndrome disrupt protein-protein interfaces observed in the recruitment state.

Structures of the human CST-Polα-primase complex bound to telomere templates

Qixiang He et al.

Summary: This study reveals that the CST accessory protein helps organize the Pol alpha-primase enzyme for efficient primer synthesis. Cryogenic electron microscopy structures show that CST separates the DNA and RNA catalytic centers of Pol alpha-primase into two domains and arranges them in the correct order for RNA-DNA synthesis. The study also provides insights into the template-binding specificity of CST and its role in the assembly and activation of the CST-Pol alpha-primase complex.

PROXIMA-1 beamline for macromolecular crystallography measurements at Synchrotron SOLEIL

Leonard M. G. Chavas et al.

Summary: The PROXIMA-1 undulator beamline at Synchrotron SOLEIL is dedicated to biomolecular crystallography experiments and has a layout designed to favor anomalous data recording and studies of crystals with large cell dimensions. Over 12 years, it has accommodated a large number of shifts and visitors, resulting in numerous published scientific papers and coordinates deposited in the Protein Data Bank. The beamline is equipped with advanced beam-shaping equipment, a three-axis goniometer, an X-ray detector, and a sample exchange robot, allowing for standard experimental schemes as well as remote or `mail-in' access.

Highly accurate protein structure prediction with AlphaFold

John Jumper et al.

Summary: Proteins are essential for life, and accurate prediction of their structures is a crucial research problem. Current experimental methods are time-consuming, highlighting the need for accurate computational approaches to address the gap in structural coverage. Despite recent progress, existing methods fall short of atomic accuracy in protein structure prediction.
