Best practices for time-resolved serial synchrotron crystallography

Eike C. Schulz et al.

Summary: With advancements in X-ray sources, equipment, and data-analysis tools, time-resolved crystallographic experiments are becoming more accessible to a wider user base. This article highlights the key factors that need to be considered when planning and conducting a time-resolved structural study, with a focus on synchrotron-based experiments.

Effects of cryo-EM cooling on structural ensembles

Lars Bock et al.

Summary: Structure determination by cryo-EM provides atomic-resolution information on structural heterogeneity and ensembles. This study focuses on quantifying the effects of cooling during cryo-EM imaging by combining continuum model calculations, molecular dynamics simulations, and kinetic models. The findings suggest that thermal contraction, reduced thermal motion, and equilibration into lower free-energy conformations contribute to the narrowing of structural ensembles. This research provides a method to quantify the heterogeneity of room-temperature ensembles from cryo-EM structures.

Millisecond mix-and-quench crystallography (MMQX) enables time-resolved studies of PEPCK with remote data collection

Jonathan A. Clinger et al.

Summary: Millisecond mix-and-quench crystallography (MMQX) has been developed to achieve millisecond time-resolved data using fewer crystals and routine remote synchrotron data collection. This approach lowers the barrier to time-resolved crystallography and enables a broad expansion in structural studies of protein dynamics.
