Whole-genome sequencing reveals host factors underlying critical COVID-19

Athanasios Kousathanas et al.

Summary: This study used whole-genome sequencing to compare the genomes of 7,491 critically ill individuals with 48,400 controls, and identified 23 independent variants associated with critical COVID-19. The research found multiple genes that are involved in interferon signaling, leukocyte differentiation, and blood-type antigen secretion to be associated with critical illness.

Regulation and Role of αE Integrin and Gut Homing Integrins in Migration and Retention of Intestinal Lymphocytes during Inflammatory Bowel Disease

Mary E. Keir et al.

Summary: Targeting interactions between alpha 4 beta 7 integrin and endothelial adhesion molecule MAdCAM-1 to inhibit lymphocyte migration to the gastrointestinal tract is effective in treating inflammatory bowel disease. Upregulation of ICAM-1, VCAM-1, and MAdCAM-1 at gene and protein levels was observed in both ileal and colonic tissue from active IBD patients compared to healthy subjects. TGF-beta 1 treatment induced alpha E expression on T cells, suggesting a potential mechanism for upregulating alpha E beta 7 in response to TGF-beta within the gut mucosa of IBD patients.

Single-cell analyses of Crohn's disease tissues reveal intestinal intraepithelial T cells heterogeneity and altered subset distributions

Natalia Jaeger et al.

Summary: Crohn's disease is a chronic transmural inflammation in the gut caused by dysregulated interaction between microbiome and gut immune system. Studies have shown that T cells from intestinal epithelium and lamina propria in CD patients exhibit unique subsets and altered phenotypes during inflammation.

Ultra-high Magnification Endocytoscopy and Molecular Markers for Defining Endoscopic and Histologic Remission in Ulcerative Colitis - An Exploratory Study to Define Deep Remission

Marietta Iacucci et al.

Summary: The study showed that endocytoscope scoring system can effectively predict histological remission in patients with ulcerative colitis. Soluble markers such as BDNF, MIP-1 alpha, and sVCAM-1 were identified to predict histological remission. RNA sequencing analysis revealed gene expressions and pathways that discriminate healed from nonhealed mucosa.

Randomized Controlled Trial Substudy of Cell-specific Mechanisms of Janus Kinase 1 Inhibition With Upadacitinib in the Crohn's Disease Intestinal Mucosa: Analysis From the CELEST Study

Daniel Aguilar et al.

Summary: This study aimed to investigate the mechanistic insights of the JAK1-selective inhibitor upadacitinib in patients with Crohn's disease, showing that upadacitinib modulates inflammatory pathways in mucosal lesions of patients with anti-TNF-refractory Crohn's disease, including the reversal of overexpression of inflammatory fibroblast and interferon-gamma effector signature markers.
