Compressed sensing for electron cryotomography and high-resolution subtomogram averaging of biological specimens

Jan Bohning et al.

Summary: Cryoelectron tomography and subtomogram averaging enable direct visualization and study of biological macromolecules in their native cellular environment. However, limitations such as low signal-to-noise ratios, low particle abundance, and low throughput have restricted the obtainable structural information. This study demonstrates the potential of a compressed sensing approach to enhance the visualization and resolution of tomograms, addressing these limitations.

HEMNMA-3D: Cryo Electron Tomography Method Based on Normal Mode Analysis to Study Continuous Conformational Variability of Macromolecular Complexes

Mohamad Harastani et al.

Summary: "Cryogenic electron tomography (cryo-ET) allows for structural determination of biomolecules in their native environment, but challenges in data analysis have hindered exploration of its potential in studying dynamics of macromolecular complexes within cells. The HEMNMA-3D method presented here addresses the continuous conformational changes of complexes in cryo electron subtomograms by combining elastic and rigid-body 3D-to-3D iterative alignments. These alignments provide insights into the distribution and dynamics of complexes, visualized in lower dimensional space."

Structural features of nucleosomes in interphase and metaphase chromosomes

Yasuhiro Arimura et al.

Summary: This study utilized a new cryo-EM pipeline to reconstruct nucleosome structures in interphase and metaphase chromosomes, revealing structural variations in linker DNA, histone tails, and nucleosome core particles. The high-resolution structures showed DNA-sequence-independent stabilization of superhelical locations and the preferential binding of linker histone H1.8 to metaphase chromatin.

In situ cryo-electron tomography reveals gradient organization of ribosome biogenesis in intact nucleoli

Philipp S. Erdmann et al.

Summary: The large and small subunits of the ribosome are synthesized independently within the nucleolus before being exported into the cytoplasm, where they assemble into functional ribosomes. Using in situ cryo-electron tomography, the researchers observed ribosome maturation and revealed the native organization of the nucleolus, providing new insights into ribosome biogenesis.
