Autophagy Blockade Limits HER2+Breast Cancer Tumorigenesis by Perturbing HER2 Trafficking and Promoting Release Via Small Extracellular Vesicles

Mingang Hao et al.

Summary: This study identified a mechanism by which disruption of FIP200-mediated autophagy can reduce HER2 expression on tumor cells and inhibit mammary tumorigenesis. Inhibition of sEVs secretion can rescue HER2 levels in tumor cells, suggesting a role for autophagy in promoting tumorigenesis in HER2+ breast cancer.

Autophagy mediated lipid catabolism facilitates glioma progression to overcome bioenergetic crisis

Chenran Wang et al.

Summary: Activation of mTORC1 is important in cancer development. This study revealed that autophagy-mediated lipid catabolism is essential to sustain mTORC1 activity in glioma cells under energy stresses. Inhibition of autophagy or FAO inhibitors with 2DG effectively suppressed the progression of xenografted glioma with hyper-activated mTORC1.
