Integrated Pipeline of Rapid Isolation and Analysis of Human Plasma Exosomes for Cancer Discrimination Based on Deep Learning of MALDI-TOF MS Fingerprints

Summary: In this study, a comprehensive strategy for rapid isolation and analysis of plasma exosomes from human blood was presented, and deep learning fingerprints of plasma exosomes were used to discriminate different cancers excellently. Sequential size-exclusion chromatography (SSEC) was employed for efficient separation of plasma exosomes, and matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MS) accurately profiled the contents encapsulated in exosomes. By constructing an Exo-ANN model, a high diagnostic accuracy for cancer was achieved in clinical samples.

Electrical characterization of tumor-derived exosomes by conductive atomic force microscopy

Summary: This study used conductive atomic force microscopy (C-AFM) to characterize the electrical properties of tumor-derived exosomes. It was found that the conductivity of exosomes was influenced by external factors and was also compared between low and high metastatic potential cell-derived exosomes. This work is important for better understanding the physical properties of tumor-derived exosomes and promoting their clinical applications.

Characterization and modulation of surface charges to enhance extracellular vesicle isolation in plasma

Summary: Extracellular vesicles (EVs) have the potential for disease diagnosis, but are often contaminated by low density lipoproteins (LDLs) when isolated from blood. By studying the difference in electric charge between EVs and LDLs, we developed a method using ion-exchange chromatography to separate EVs from LDLs. This finding improved the analytical signals in EV molecular assays.

Comparison of Human Urinary Exosomes Isolated via Ultracentrifugation Alone versus Ultracentrifugation Followed by SEC Column-Purification

Summary: This study compared two methods, ultracentrifugation and size exclusion chromatography, for isolating exosomes from urine. Multiplexed single-cell analysis of FNA allows accurate diagnosis of salivary gland tumors

Summary: The authors developed a multiplexed cycling technology for the rapid analysis of single cells in salivary gland tumors, allowing for molecular analysis and diagnosis. They validated antibodies against 29 protein markers associated with different subtypes of salivary gland tumors, and developed a FAST molecular diagnostic assay that can distinguish between benign and malignant tumors. Using multiple markers increased the diagnostic accuracy compared to using a single marker.

Sequencing-Based Protein Analysis of Single Extracellular Vesicles
