Summary: The discovery of novel antibacterials without cross resistance is crucial, but suitable pathways to target are limited. At Actelion Pharmaceuticals, we focused on researching novel bacterial topoisomerase inhibitors (NBTI) to find new broad spectrum antibiotics with limited resistance for severe hospital infections. This paper summarizes our efforts, resulting in the discovery of chemical classes with potent Gram-negative activity and several compounds undergoing preclinical profiling.
