Discovery of Inhibitors of DNA Methyltransferase 2, an Epitranscriptomic Modulator and Potential Target for Cancer Treatment

Marvin Schwickert et al.

Summary: Selective manipulation of the epitranscriptome through the design and analysis of inhibitors for the tRNA methyltransferase DNMT2 could be beneficial for cancer treatment and expand our understanding of epigenetic inheritance. New synthetic inhibitors with similar binding and inhibitory potencies as natural compounds were discovered. However, there are limitations in cellular activity.

An Optimized Microscale Thermophoresis Method for High- Throughput Screening of DNA Methyltransferase 2 Ligands

Robert Alexander Zimmermann et al.

Summary: This study explores the applicability of a novel fluorescent tracer for human RNA methyltransferase DNMT2 and establishes a new screening method using microscale thermophoresis. The results overcome some limitations of traditional methods and provide additional information.

Diversity of the reaction mechanisms of SAM-dependent enzymes

Qiu Sun et al.

Summary: This article mainly describes the importance of non-methylation reactions of SAM-dependent enzymes in biosynthesis, compares the structural and mechanistic similarities and distinctions between SAM-dependent MTases and non-methylating SAM-dependent enzymes, summarizes the reactions catalyzed by these enzymes, and discusses the structural conservation and catalytical diversity of class I-like non-methylating SAM-dependent enzymes. It also proposes a possibility in enzyme evolution and suggests future perspectives for enzyme-mediated chemistry and biotechnology for the development of new methods for drug synthesis.

Sperm microRNAs confer depression susceptibility to offspring

Yanbo Wang et al.

Summary: The findings demonstrate that parental life experiences can influence offspring traits through epigenetic inheritance, shedding light on the etiology of depression. It is shown that F1 offspring born to depression-like model males exhibit depression-like symptoms at molecular, neuronal, and behavioral levels. The study reveals that sperm miRNAs can reshape early embryonic transcriptional profiles in core neuronal circuits towards depression-like phenotypes, providing insight into the mechanism underlying susceptibility to depression.

The Promise and Peril of Chemical Probe Negative Controls

Jinyoung Lee et al.

Summary: The study compared the selectivity profiles of four chemical probes and their respective negative controls, finding that chemical modifications neutralizing the charge can result in up to 80% of known off-targets being inactive against the control. Careful selection of negative controls for chemical probes is emphasized, and it is best practice to verify that two unrelated chemical probes targeting the same protein elicit the same phenotype.
