Peroxide Antimalarial Drugs Target Redox Homeostasis in Plasmodium falciparum Infected Red Blood Cells

Summary: This study used chemoproteomics to investigate the protein alkylation targets of peroxide antimalarials and found a significant alkylation of proteins involved in redox homeostasis. Disrupted redox processes were confirmed and changes in cellular thiol levels were observed. These findings reveal the mechanism of action of peroxide antimalarials.

Summary: Recent research has highlighted the need for fully synthetic endoperoxide-based antimalarials due to the rise of resistance to artemisinin in Southeast Asia and the synthetic limitations of the artemisinin scaffold. Various synthetic endoperoxide antimalarials utilizing different endoperoxide warheads have been explored, with two classes - 1,2,4-trioxolanes and 1,2,4,5-tetraoxanes - showing promise and still actively being developed.

Enantioselective Synthesis and Profiling of Potent, Nonlinear Analogues of Antimalarial Tetraoxanes E209 and N205

Summary: Research has developed a method for synthesizing nonlinear analogues of second-generation antimalarials, showing up to 10-fold improvement in solubility compared to linear counterparts. Pharmacokinetic studies in rats revealed that a selected nonlinear analogue demonstrated significantly improved oral absorption and exposure in vivo, with more than double the AUC and increased oral bioavailability.
