Identification of a small molecule splicing inhibitor targeting UHM domains

Asaki Kobayashi et al.

Summary: Splicing factor mutations are common in various cancers, and cells with these mutations are vulnerable to certain drugs. Through virtual screening and experimental validation, researchers discovered a small molecule, UHMCP1, that can disrupt specific splicing factor interactions, affecting RNA splicing and cell viability.

Complex landscape of alternative splicing in myeloid neoplasms

Courtney E. Hershberger et al.

Summary: Myeloid neoplasms exhibit frequent mutations in spliceosome components, resulting in distinct abnormal splicing signatures with little overlap. RNA-Seq analysis on a large cohort identified commonly mis-spliced transcripts and revealed the impact of splicing factor mutations on protein function.Meta-splicing analysis showed reduced levels of retained introns in disease samples, exacerbated in patients with splicing factor mutations, suggesting a common pathway by which alternative splicing may regulate cell proliferation.

U2af1 is a haplo-essential gene required for hematopoietic cancer cell survival in mice

Brian A. Wadugu et al.

Summary: Research indicates that deletion of the wild-type allele in hematopoietic cells expressing U2AF1(S34F) heterozygous mutation is lethal, confirming the dependence of mutant U2AF1 cells on the expression of wild-type U2AF1 for survival. Leukemia cells carrying mutant U2AF1 are more sensitive to reduced wild-type U2AF1 expression, suggesting a potential therapeutic strategy for selectively targeting the wild-type U2AF1 allele in mutant cells.

A synthetic small molecule stalls pre-mRNA splicing by promoting an early-stage U2AF2-RNA complex

Rakesh Chatrikhi et al.

Summary: This study identifies a compound that enhances RNA binding by the U2AF2 subunit and inhibits splicing of representative substrates, effectively killing cancer cells by stalling spliceosome assembly. The results suggest that trapping early spliceosome assembly could be an effective pharmacological approach for manipulating pre-mRNA splicing.
