CircRNA_001895 promotes sunitinib resistance of renal cell carcinoma through regulation of apoptosis and DNA damage repair

Lei Tan et al.

Summary: This study found that the expression of circRNA_001895 was higher in sunitinib-resistant RCC tissues and cells. Knockdown of circRNA_001895 suppressed cell proliferation and promoted cell apoptosis, while affecting the DNA damage repair mechanism. These findings suggest that circRNA_001895 may be a potential therapeutic target for advanced RCC.

circFARP1 enables cancer-associated fibroblasts to promote gemcitabine resistance in pancreatic cancer via the LIF/STAT3 axis

Chonghui Hu et al.

Summary: This study found that the CAF-specific circRNA circFARP1 plays a critical role in pancreatic ductal adenocarcinoma (PDAC), and is associated with gemcitabine resistance and poor patient survival. Mechanistically, circFARP1 regulates the interaction between CAV1, ZNRF1, and LIF, influencing the development of tumor cell stemness and drug resistance. Additionally, high levels of circFARP1 are associated with elevated serum LIF levels and poor patient survival in PDAC.

Targeting HNRNPM Inhibits Cancer Stemness and Enhances Antitumor Immunity in Wnt-activated Hepatocellular Carcinoma

Gui-Qi Zhu et al.

Summary: This study reveals the role of HNRNPM in regulating the positive association between cancer stemness and immune evasion in hepatocellular carcinoma. By controlling the alternative splicing of MBD2, HNRNPM affects the activation of the WNT/beta-catenin pathway and further regulates the expression of OCT4 and SOX2. Inhibition of HNRNPM promotes the activation of CD8+ T cells and enhances the effectiveness of anti-PD-1 immunotherapy. These findings provide new insights for the development of tailored immunotherapeutic approaches for hepatocellular carcinoma.

Summary: This study reveals that overexpression of RNF7 in renal cell carcinoma is associated with poor prognosis. RNF7 overexpression activates the JAK/STAT3 signaling pathway and inhibits SOCS1 protein, promoting glycolysis and inhibiting apoptosis in RCC cells. It also affects the sensitivity of RCC cells to sunitinib.

Inhibition of EGFR Overcomes Acquired Lenvatinib Resistance Driven by STAT3-ABCB1 Signaling in Hepatocellular Carcinoma

Beiyuan Hu et al.

Summary: Lenvatinib, a first-line treatment for hepatocellular carcinoma (HCC), has limited clinical benefits. This study reveals that HCC cells develop resistance to Lenvatinib by activating the EGFR-STAT3-ABCB1 pathway. Combined treatment with erlotinib, an EGFR inhibitor, may overcome acquired resistance and improve the clinical benefit of Lenvatinib.

Circular RNA circFIRRE drives osteosarcoma progression and metastasis through tumorigenic-angiogenic coupling

Lingfeng Yu et al.

Summary: In this study, circFIRRE was identified as an upregulated circular RNA in osteosarcoma, and its aberrant expression was associated with metastasis and poor prognosis. CircFIRRE promoted osteosarcoma progression and lung metastasis by inducing both tumor cells and blood vessels, through sponging miR-486-3p and miR-1225-5p. These findings suggest that circFIRRE could serve as a novel prognostic biomarker and therapeutic target for refractory osteosarcoma.

Blockade of STAT3/IL-4 overcomes EGFR T790M-cis-L792F-induced resistance to osimertinib via suppressing M2 macrophages polarization

Yiting Sun et al.

Summary: This study investigates the mechanism of resistance to osimertinib in non-small cell lung cancer patients with EGFR L792F mutation. The results show that EGFR L792F mutation is associated with decreased sensitivity to osimertinib and enhanced M2 polarization of macrophages. Further analysis reveals that the EGFR T790M-cis-L792F mutation upregulates phosphorylation of STAT3 Tyr705, leading to increased IL-4 expression and M2 polarization. Targeting the STAT3/IL-4 pathway could be a potential strategy to overcome osimertinib resistance.
