L-OPA1 deficiency aggravates necroptosis of alveolar epithelial cells through impairing mitochondrial function during acute lung injury in mice

Hui-Ling Jiang et al.

Summary: This study investigated the role of L-OPA1 deficiency-mediated mitochondrial dysfunction in the necroptosis of alveolar epithelial cells (AECs). The findings revealed that inhibition of L-OPA1 aggravated the pathological injury, inflammatory response, and necroptosis in the lungs of LPS-induced acute lung injury mice. In vitro experiments demonstrated that inhibition of L-OPA1 induced necroptosis of AECs, while activation of L-OPA1 alleviated necroptosis of AECs under the LPS challenge.

Opa1 and Drp1 reciprocally regulate cristae morphology, ETC function, and NAD plus regeneration in KRas-mutant lung adenocarcinoma

Dane T. Sessions et al.

Summary: Oncogenic KRas activates mitochondrial fission through phosphorylation of Drp1. In lung adenocarcinoma driven by KRas, deletion of Drp1 has no effect on tumor growth, while deletion of Opa1 inhibits tumor growth by disrupting cristae morphology and inhibiting electron transport chain activity.

Mitochondrial oxidative metabolism contributes to a cancer stem cell phenotype in cholangiocarcinoma

Chiara Raggi et al.

Summary: The study reveals that cancer stem cells in cholangiocarcinoma have a more efficient respiratory phenotype, relying on mitochondrial oxidative metabolism and PGC-1a to maintain CSC features. Targeting mitochondrial complex I or PGC-1a in CCA-SPH impairs spherogenicity and expression of markers related to CSC phenotype, pluripotency, and epithelial-mesenchymal transition. Administration of metformin or SR-18292 in mice with CCA-SPH xenografts significantly reduces tumor growth and reverts the phenotype to that of cells grown in monolayer.

Drug resistance of targeted therapy for advanced non-small cell lung cancer harbored EGFR mutation: from mechanism analysis to clinical strategy

Yuxin Zhao et al.

Summary: The mechanisms of EGFR-TKIs resistance in NSCLC include EGFR mutations, upregulation of HER2, HGF/c-MET, VEGF, IGF1, EMT and STAT3 pathways, mutations of PTEN, RAS and BRAF genes, and activation of other bypass pathways. These interconnected mechanisms can be potential targets for the treatment of NSCLC.

Suntae Kim et al.

Summary: This study identified different metabolic properties in EGFR-TKI resistant lung cancer cells compared to their parental cells, suggesting vulnerability to OXPHOS inhibitors like phenformin. Targeting mitochondrial complex 1 may be an effective therapeutic option for NSCLC with acquired EGFR-TKI resistance.

The mitochondrial protein Opa1 promotes adipocyte browning that is dependent on urea cycle metabolites

Camilla Bean et al.

Summary: The study demonstrates that the mitochondrial cristae biogenesis protein Opa1 plays a role in cell-autonomous adipocyte browning through the regulation of the urea cycle. The findings suggest a potential therapeutic strategy for obesity and highlight the connection between the urea cycle and adipocyte browning.
