Structure of monkeypox virus DNA polymerase holoenzyme

Qi Peng et al.

Summary: The World Health Organization declared monkeypox a global public health emergency in July 2022 and urgent measures are needed. Using cryo-electron microscopy, the structure of the monkeypox virus polymerase holoenzyme in complex with DNA was determined. It was found that the holoenzyme utilizes a forward sliding clamp processivity mechanism for viral DNA replication, similar to other B-family DNA polymerases. These findings provide insight into the replication mechanism of the monkeypox virus and may aid in the development of anti-poxvirus drugs.

The logic of virus evolution

Eugene Koonin et al.

Summary: Viruses are parasites that rely on host cells, but they have their own genomes and independent evolutionary trajectories. Virus genes responsible for genome replication descended from primordial replicators, while most other genes were acquired from hosts. Host gene capture occurs through two main routes: recruitment of genes that benefit virus reproduction and repurposing of host proteins for unique virus functions. This leads to varying levels of similarity between virus and host homologs.

Dissecting the roles of Haspin and VRK1 in histone H3 phosphorylation during mitosis

Summary: This study investigates the role of protein kinases in the phosphorylation of histones during cell division. The researchers find that Haspin plays a crucial role in mitosis, but the significance of VRK1 activity in this process remains unclear. A vast pool of lineage-specific microproteins encoded by long non-coding RNAs in plants

Igor Fesenko et al.

Summary: Pervasive transcription of eukaryotic genomes results in the expression of long non-coding RNAs (lncRNAs), with many potentially encoding small open reading frames (smORFs) that can be translated into peptides or microproteins. A comprehensive analysis of lncRNAs-smORFs from a moss species across 479 plant transcriptomes revealed that while many smORFs are subject to purifying selection, they represent a pool for functional innovation. Proteomic analysis confirmed translation of novel species-specific smORFs and indicated that microproteins encoded by smORFs are a functionally diverse component of the plant proteome.

Ubiquitin ligation to F-box protein targets by SCF-RBR E3-E3 super-assembly

Summary: E3 ligases are typically classified by hallmark domains such as RING and RBR. However, many neddylated CRLs and ARIH family E3 ligases form E3-E3 super-assemblies, which account for nearly half of all ubiquitin ligases in humans. By using activity-based chemical probes and cryo-electron microscopy, it has been shown that the E3-E3 mechanism can ubiquitylate diverse substrates, providing insights into the widespread ubiquitylation mechanism.

Beate Bersch et al.

Summary: Poxviruses are enveloped viruses with a linear, double-stranded DNA genome, and viral DNA synthesis is achieved by a DNA polymerase holoenzyme composed of three essential proteins. The catalytic subunit E9 forms a complex with the heterodimeric processivity factor made up of D4 and A20, which have a specific interaction involving a poxvirus-specific insertion. The interface between E9 and A20 comprises conserved hydrophobic residues and has been structurally characterized, providing insights into their molecular interactions.

Mutation Rates, Mutation Frequencies, and Proofreading-Repair Activities in RNA Virus Genetics

Esteban Domingo et al.

Summary: The error rate during template copying for viral RNA progeny production is a biologically relevant parameter for virus replication complexes, with implications for virus-host interactions and virus diversification. Mutation rates for RNA viruses range from 10(-3) to 10(-6) copying errors per nucleotide incorporated into the new RNA product. Some viruses possess error correction mechanisms, like the proofreading-repair 3' to 5' exonuclease in coronaviruses, to decrease error rates during replication and maintain genetic stability.

Ancient Gene Capture and Recent Gene Loss Shape the Evolution of Orthopoxvirus-Host Interaction Genes

Tatiana G. Senkevich et al.

Summary: The survival of viruses relies on their ability to resist host defenses and certain genes play a crucial role in this process. Poxviruses, like Orthopoxviruses (ORPV), have a significant number of genes dedicated to counteracting host immunity. Through evolution, ORPV acquired accessory genes in three major waves, leading to gene duplication and the formation of gene families. Interestingly, most accessory genes were lost during ORPV evolution, suggesting a variety of functional interactions within this virus family.
