Summary: Inorganic polyphosphate is an ancient and ubiquitous polymer that is present in all organisms. While its role in bacteria and yeast has been described, its exact function in mammalian physiology remains poorly understood. Our study suggests that the metabolism of mitochondrial polyphosphate could be a potential pharmacological target in neurodegenerative disorders and neuronal aging.

Summary: This review summarizes recent insights into the integration of phosphate metabolism with mechanisms of fungal adaptation, highlighting the importance of phosphate sensing and regulation in fungal pathogenesis, host immune responses, and disease outcomes. New studies also provide insights into the influence of phosphate availability on interactions between fungi and other microbes, mycorrhizal and endophytic associations with plants, and connections with plant immunity. Phosphate homeostasis is emerging as an integral part of fungal metabolism and communication to support diverse lifestyles.

The Cryptococcus neoformans Flc1 Homologue Controls Calcium Homeostasis and Confers Fungal Pathogenicity in the Infected Hosts

Summary: This study reveals the importance of the Flc1 protein in maintaining calcium homeostasis and vacuolar biogenesis in Cryptococcus neoformans. Deletion of Flc1 leads to an increase in cytosolic calcium concentration, an elevation of Crz1 transcription factor in the nucleus, and an aberrant accumulation of cell wall chitin. Flc1 is also involved in vacuolar fusion and capsule formation. The absence of Flc1 results in the loss of survival ability and virulence in Cryptococcus.

Summary: This study explores how the InsP(8)-SPX complex regulates plant Pi homeostasis by targeting the unique CC domain of PHRs. Structural analysis reveals that mutations in the CC domain affect the DNA binding capability of PHRs, resulting in constitutive Pi starvation responses.
