Chimeric Human Papillomavirus-16 Virus-like Particles Presenting P18I10 and T20 Peptides from HIV-1 Envelope Induce HPV16 and HIV-1-Specific Humoral and T Cell-Mediated Immunity in BALB/c Mice

Summary: In this study, chimeric HPV:HIV VLPs were constructed by inserting HIV-1 P18I10 CTL peptide and T20 anti-fusion peptide into the HPV16 L1 capsid protein. The insertion did not affect the stability, self-assembly, morphology, and antibody reactivity of the VLPs. The chimeric VLPs induced both HPV16- and HIV-1-specific immune responses, making them a potential vaccine platform for controlling HPV16 and HIV-1 infection.

The impact of size on particle drainage dynamics and antibody response

Summary: In this study, engineered CCMVTT-VLPs were investigated for their draining kinetics and immunogenicity, with Round-shaped CCMVTT-VLPs showing more efficient drainage to secondary lymphoid organs and greater systemic IgG and IgA responses compared to Rod-shaped CCMVTT-VLPs. Additionally, Round-shaped CCMVTT-VLPs led to prominent formation of splenic germinal centers and polarization of T cell response towards Th1. This is the first study to compare the draining kinetics and immunogenicity of VLPs forming nano-sized icosahedra or micrometer-sized rods.

Summary: The VP60 capsid protein from rabbit haemorrhagic disease virus can form virus-like particles when expressed using different protein expression systems. In this study, a dual-serving RHDV vaccine was successfully developed using CrisBio technology, which allows the co-expression of VP60 proteins from two RHDV serotypes. Rabbits immunised with the chimeric VLPs were fully protected against a lethal challenge infection with the two RHDV serotypes, demonstrating the effectiveness of the dual vaccine strategy.
