T follicular helper cells and their impact on humoral responses during pathogen and vaccine challenge

Jennifer A. Juno et al.

Summary: T follicular helper (Tfh) cells play a crucial role in the development, maintenance, and output of the germinal centre (GC) response, impacting vaccine immunity. Different cytokine environments influence Tfh cell differentiation, while sampling lymph nodes offers new avenues for studying GC responses directly.

Influenza vaccines: where we are, where we are going

Nadim Khalil et al.

Summary: Influenza vaccines are the most effective strategy for preventing influenza illness, but currently available vaccines have suboptimal efficacy. Therefore, further research and innovation are needed to develop a universal influenza vaccine that provides long-lasting immunity and protects against a broad range of strains.

Vaccines elicit highly conserved cellular immunity to SARS-CoV-2 Omicron

Jinyan Liu et al.

Summary: This study demonstrates that cellular immunity induced by current SARS-CoV-2 vaccines is highly conserved to the Omicron spike protein. Individuals vaccinated with Ad26.COV2.S or BNT162b2 vaccines showed durable spike-specific CD8(+) and CD4(+) T cell responses that were cross-reactive to both the Delta and Omicron variants, including in central and effector memory cellular subpopulations.

Development of a Mouse Model to Explore CD4 T Cell Specificity, Phenotype, and Recruitment to the Lung after Influenza B Infection

Ajitanuj Rattan et al.

Summary: In this study, a mouse model of intranasal infection with influenza B virus was developed, and a series of robustly elicited individual CD4 T cell peptide specificities were identified. The CD4 T cells were localized to both lung vasculature and tissue, and were highly enriched in lung tissue after infection. The CD4 T cells expressed prototypical markers of tissue residency. These studies will enable more sophisticated analyses of influenza B virus infection.

CD4+ Cytotoxic T cells - Phenotype, Function and Transcriptional Networks Controlling Their Differentiation Pathways

Teresa Preglej et al.

Summary: CD4 CTLs are a subset of T cells that display cytotoxic activity and are associated with viral infections and cancer immunity. They may also contribute to immunopathology. The molecular mechanisms regulating CD4 CTL differentiation are poorly understood, with recent data suggesting a role for histone deacetylases in their generation.

Tissue resident memory T cells- A new benchmark for the induction of vaccine-induced mucosal immunity

Mariah Hassert et al.

Summary: Historically, the induction of neutralizing antibodies in the blood has been the gold-standard benchmark for vaccine immunogenicity. However, there is now growing recognition of the importance of mucosal immunity and tissue resident memory T cells (T-rm) in the early detection and restriction of mucosal pathogens.

Potentiating Lung Mucosal Immunity Through Intranasal Vaccination

Sean A. Nelson et al.

Summary: Administration of yearly influenza vaccines is the most effective way to control the spread of influenza virus, but practical and immunological factors can lead to sub-optimal vaccine efficacy. The development of improved or universal influenza vaccines has been a focus within the field, with attention on the route of immunization to elicit tissue resident memory populations. The unique features of mucosal immunity in the upper and lower respiratory tracts suggest the importance of antigen localization in intranasal vaccine design for eliciting protective B and T cell immunity.

Development of combination adjuvant for efficient T cell and antibody response induction against protein antigen

Yasunari Haseda et al.

Summary: The DOTAP/D35/aluminum salt adjuvant developed in this study induced strong T cell and antibody responses against a model protein antigen with a single immunization. In vitro studies showed that strong mutual interactions among the adjuvant component and the model antigen were important factors for this efficient immune induction. In vivo imaging of the antigen distribution indicated that the DOTAP component in the adjuvant formulation elicited transient antigen accumulation at draining lymph nodes, possibly by antigen uptake DC migration.

Targeting Antigens for Universal Influenza Vaccine Development

Quyen-Thi Nguyen et al.

Summary: Traditional influenza vaccines are strain-specific and require annual reformulation, whereas a universal influenza vaccine is critical for long-term protection and potential pandemics. Different target antigens for UIVs have advantages and limitations in generating immune responses against divergent influenza viruses.
