Inhibition of interferon I induction by non-structural protein NSs of Puumala virus and other vole-associated orthohantaviruses: phenotypic plasticity of the protein and potential functional domains

Summary: The study investigated the inhibitory activity of the NSs proteins of Puumala virus and other vole-borne hantaviruses, showing that translation initiation at different start codons can influence their inhibitory activity. It was found that amino acid variations in the NSs protein are well tolerated, indicating its phenotypic plasticity. Additionally, the N-terminal 20-amino-acid region of the NSs protein was identified to be associated with strong inhibition and highly sensitive to amino acid exchanges and tag fusions.

Summary: Research using CRISPR/Cas9 technology to knockout candidate hantavirus receptors in human endothelial cells found that the loss of beta 3 integrin, beta 1 integrin, and/or DAF had little or no effect on hantavirus entry, while loss of protocadherin-1 significantly reduced virus entry and infection. Major host molecules necessary for hantavirus entry by PCDH1-independent mechanisms are yet to be discovered.
