The researchers found that isolates with increased Olorofim minimum inhibitory concentrations (MICs) could be selected under laboratory conditions. Gene sequencing revealed amino acid substitutions in the PyrE gene of these resistant isolates, which decreased the affinity of Olorofim to the target protein dihydroorotate dehydrogenase (DHODH). Additionally, certain mutations were found to reduce the growth rate of the fungus.
