Protein-metabolite interactomics of carbohydrate metabolism reveal regulation of lactate dehydrogenase

Summary: Metabolic networks play a crucial role in various cellular processes. Discovering low-affinity protein-metabolite interactions that govern these networks is challenging. A new method, called mass spectrometry integrated with equilibrium dialysis for the discovery of allostery systematically (MIDAS), was developed to identify such interactions. In a study involving enzymes from human carbohydrate metabolism, 830 protein-metabolite interactions were discovered, including both known and unknown interactions. Some of these interactions were functionally validated, revealing the isoform-specific inhibition of lactate dehydrogenase by long-chain acyl-coenzyme A. The findings suggest that these protein-metabolite interactions contribute to the adaptive metabolic flexibility in different tissues.

Deep learning algorithms have emerged as a promising method in multi-omics data analysis due to their predictive performance and ability to capture nonlinear and hierarchical features. However, integrating and translating multi-omics data into functional insights remains a challenge, but there is a clear trend towards incorporating multi-omics analysis in biomedical research to explain complex relationships between molecular layers.

Metabolic determination of cell fate through selective inheritance of mitochondria

This metabolic feature, established by functionally distinct mitochondrial age-classes, acts as a fate determinant in asymmetric division of epithelial stem-like cells.Cells that inherit old mitochondria with oxidative energy metabolism promote differentiation, while cells inheriting newly synthesized mitochondria with low levels of Rieske iron-sulfur polypeptide 1 maintain stemness through the pentose phosphate pathway during early fate determination.

Defining mitochondrial protein functions through deep multiomic profiling

Summary: Mitochondria, as the epicentres of eukaryotic metabolism and bioenergetics, are linked to numerous diseases. However, the functions of many mitochondrial proteins are still unclear, and the genetic basis of many mitochondrial disorders remains unresolved. Through mass spectrometry-based multiomics analyses, we have established a comprehensive functional compendium of human mitochondrial proteins, providing a foundation for mechanistic investigations and genetic diagnosis of mitochondrial diseases. Our findings have identified specific proteins associated with mitochondrial function and disorders, and defined a rich cell signature of mitochondrial dysfunction.
