A transcriptomic study on human lymphoblastic T cells revealed altered gene expressions caused by low doses of AFB1, OTA, and their combination. Significant differential gene expressions were observed in all conditions, with immune system-related processes and functions affected. Pathway analysis indicated potential DNA damage, metabolic reprogramming, and cell death with oxidative stress as mechanisms of immune toxicity.

Protective role of fermented whey and pumpkin extract against aflatoxin B1 and ochratoxin A toxicity in Jurkat T-cells

Summary: The beneficial effect of fermented whey and pumpkin extract rich in carotenoids was assessed in Jurkat cells exposed to aflatoxin B-1 and ochratoxin A. Proteomic analysis showed that these functional ingredients were able to mitigate the cytotoxicity of the mycotoxins by affecting various biological processes and pathways, including antioxidant activity, nucleosome assembly, and cell growth regulation. Key proteins involved in antioxidant activity and nucleosome assembly were upregulated, while a protein associated with carcinogenic cell growth was downregulated. These findings suggest that functional ingredients can protect against genomic stress and inhibit the growth of carcinogenic cells.

Summary: Due to globalization, mycotoxins have become a major food contaminant with significant risks to human health. This review focuses on the toxic effects of AFB1 and OTA, two of the most toxic and studied mycotoxins, on human health using flow cytometry and immunofluorescence techniques in vitro and in vivo. The majority of the selected reports examine immunotoxicity, while others explore nephrotoxicity, hepatotoxicity, gastrointestinal toxicity, neurotoxicity, embryotoxicity, reproductive system, breast, esophageal and lung toxicity. Immunofluorescence analysis reveals processes related to inflammation, neuronal differentiation, DNA damage, oxidative stress, and cell death caused by AFB1 and OTA toxicity. Flow cytometry analysis includes apoptosis assay, cell cycle analysis, and intracellular ROS measurement. Further research is needed to elucidate the mechanisms of action of AFB1 and OTA on human health.

Occupational exposure to AFB1 should be given more attention in order to reduce the incidence of cancer.

Summary: The presence of mycotoxins in cereals and cereal products is a significant issue. This study evaluated the bioaccessibility of aflatoxin B-1 (AFB1) and ochratoxin (OTA) in bread and investigated the effects of milk whey and pumpkin in reducing mycotoxins bioaccessibility. The results showed that pumpkin powder was the most effective in reducing AFB1 and OTA bioaccessibility, followed by whey, fermented whey, and the combination of pumpkin-fermented whey. These findings suggest that pumpkin and milk whey could be potential bioactive ingredients for the bakery industry.

Pumpkin extract and fermented whey individually and in combination alleviated AFB1-and OTA-induced alterations on neuronal differentiation in vitro

Summary: Food and feed are often contaminated with mycotoxins, but their effects can be mitigated by functional compounds. AFB1 and OTA are the most toxic mycotoxins, and their neurotoxicity is not well studied. This study used SH-SY5Y human neuroblastoma cells to investigate the effects of pumpkin and fermented whey extracts in digested bread, along with AFB1 and OTA, on neuronal differentiation. The results showed that OTA had the most damaging effect on cell differentiation. Cell cycle analysis revealed significant differences in the G0/G1 phase for OTA-contained bread. Gene expression analysis indicated that the addition of functional ingredients in bread improved neuronal marker expression and overall gene expression compromised by mycotoxins.

Furthermore, LGG supplementation can modulate the composition of intestinal microbiota by increasing the relative abundances of beneficial bacteria and reducing the relative abundances of harmful bacteria. These findings provide a theoretical basis for the application of LGG in alleviating the adverse effects induced by DON exposure.

The importance of transcriptomics and proteomics for studying molecular mechanisms of mycotoxin exposure: A review

Summary: This review highlights recent advances in using transcriptomics and proteomics as key tools to understand the molecular toxicity of mycotoxins. The most studied mycotoxin using transcriptomics is deoxynivalenol (DON), followed by aflatoxins (AFs) and zearalenone (ZEA). In contrast, proteomics mostly focuses on AFs, with some research on ZEA and DON. However, fewer studies have investigated the toxicological effects of emerging mycotoxins such as patulin, ochratoxin A, T-2 toxin, alternariol, and amino-14,16-dimethyloctadecan-3-ol. Transcriptomics commonly studies changes in gene expression related to the immune system, while proteomics focuses on cellular processes. The most commonly used techniques are RT-qPCR for gene expression analysis and liquid chromatography coupled with mass spectrometry for proteomics assays. The data gathered indicate an increased interest in using these omic approaches in the past five years. However, in vitro models are prioritized over in vivo and ex vivo models. Therefore, there is a need to enhance the use of in vivo models and alternative methods to better understand the mode of action of mycotoxins on animal and human health.
