Current androgen receptor antagonists under investigation for resistant prostate cancer

Maria Concetta Nigro et al.

Summary: The use of androgen receptor inhibitors and taxane-based chemotherapy remains the gold standard for treating advanced prostate cancer. However, new agents like enzalutamide, apalutamide, darolutamide, and abiraterone have been approved and are being used in clinical practice. Advances in research on castration-resistant prostate cancer, including new drug development and combination therapies, are improving survival outcomes for patients.

Characterisation and validation of an in vitro transactivation assay based on the 22Rv1/MMTV_GR-KO cell line to detect human androgen receptor agonists and antagonists

Yooheon Park et al.

Summary: This study introduced and validated an androgen receptor (AR) transactivation assay for the detection of AR agonists and antagonists using a stably transfected human prostate cancer cell line. The assay showed high concordance in chemical classifications and stability across different laboratories, with accurately predicted results.

Reliability of the AR-CALUX® In Vitro Method Used to Detect Chemicals with (Anti)Androgen Activity: Results of an International Ring Trial

Anne Milcamps et al.

Summary: The AR-CALUX (R) in vitro method is an effective way to detect endocrine active chemicals with androgenic or anti-androgenic potential. An international ring trial confirmed its high reproducibility and low variability within and between laboratories, leading to almost 100% concordance of classifications. The method has been approved for inclusion in the updated OECD test guideline TG458, along with two other mechanistically similar methods.
