Regeneration Using Adipose-Derived Stem Cell Sheets in a Rabbit Meniscal Defect Model Improves Tensile Strength and Load Distribution Function of the Meniscus at 12 Weeks

Summary: The mechanical properties of meniscus tissue regenerated using adipose-derived stem cell (ADSC) sheets in a rabbit meniscal defect model were evaluated. ADSC sheets were prepared and transplanted into one knee while the contralateral knee served as a control. Mechanical tests were conducted at 4 and 12 weeks post-transplantation. The ADSC-treated knees showed higher elastic modulus and ultimate tensile strength, larger contact area, and lower peak pressure compared to the control knees. These results suggest that ADSC sheet transplantation can effectively promote meniscus regeneration.

The appropriate concentration of hypoxia enhanced the proliferation and stemness of PBMSCs, and increased the multidirectional differentiation potential.

Summary: This study explored the feasibility and repair effect of peripheral blood-derived mesenchymal stem cells (PBMSCs) combined with demineralized cortical bone matrix (DCBM) as a tissue-engineered meniscus graft. PBMSCs were found to be suitable seed cells for tissue-engineered meniscus, and the PBMSCs/DCBM composite showed the ability to form meniscus in vitro. In vivo experiments demonstrated that the PBMSCs/DCBM group had better repair effect and cartilage protection than the control groups.

This approach not only restored the anisotropy of the native healthy meniscus, but also maintained joint function and prevented secondary joint degeneration, offering a new strategy for treating meniscus injury-related joint degenerative diseases.
