Atomically Precise Water-Soluble Graphene Quantum Dot for Cancer Sonodynamic Therapy

Summary: A water-soluble graphene quantum dot with deep-red emission and excellent sonodynamic sensitization has been synthesized. It can be dispersed in water without aggregation by decorating water-soluble functional groups. The deep-red emission allows for tracking its bio-process, showing good cell-uptake and long-term retention in tumor tissue. Compared to traditional sonosensitizers, it generates superior reactive oxygen species and exhibits excellent tumor inhibition potency. The biosafety of this graphene quantum dot has been validated in both in vitro and in vivo assays.

Oxygen-carrying microfluidic microcapsules for enhancing chemo-sonodynamic therapy on patient-derived tumor organoid models

Summary: A novel microcapsule delivery system with a perfluorocarbon core carrying oxygen and a hydrogel shell encapsulating an antimetabolite and a sonosensitizer has been developed to enhance the efficacy of chemo-sonodynamic therapy against pancreatic cancer. The system is able to reverse the hypoxic microenvironment, enhance reactive oxygen species production during therapy, and induce cytotoxicity with assistance from the release of the antimetabolite in the presence of low intensity ultrasound. Significant anti-tumor effects have been achieved in patient-derived pancreatic cancer organoid models.

Engineering Cu-CuFe2O4 nanoenzyme for hypoxia-relief and GSH-depletion enhanced chemodynamic/sonodynamic therapy

Summary: In this study, a novel Cu-CuFe2O4 nanoenzyme was developed for simultaneous relief of hypoxia and depletion of GSH, enhancing the therapeutic effect of ROS-involved therapy. The nanoenzyme exhibited catalytic activities to continuously generate oxygen from tumor-overexpressed hydrogen peroxide, facilitating hypoxia-relieved sonodynamic therapy. Additionally, the Cu-CuFe2O4 NPs reacted with GSH to generate abundant hydroxyl radicals for chemodynamic therapy. The efficient anticancer ability of Cu-CuFe2O4 NPs was demonstrated in MCF-7 cell killing and multicellular tumor spheroids elimination.

Defect Engineering Triggers Exceptional Sonodynamic Activity of Manganese Oxide Nanoparticles for Cancer Therapy

Summary: In this study, oxygen-deficient manganese oxide (MnOx) nanoparticles were developed as an advanced sonosensitizer for the treatment of cancer. The introduction of oxygen defects greatly enhanced the sonodynamic activity of MnOx and its biocompatibility. The nanoparticles exhibited higher ROS production and tumor killing efficiency compared to their counterparts. Furthermore, the nanoparticles induced tumor cell death by targeting polyunsaturated fatty acids in the cell membrane with ultrasound-triggered ROS. The designed sonosensitizers also showed negligible toxicity in treated mice.
