Side chain engineering of semiconducting polymers for improved NIR-II fluorescence imaging and photothermal therapy

Chao Yin et al.

Summary: Efficient agents for NIR-II fluorescence imaging in biomedicine and life science have been developed through side chain engineering of semiconducting polymers (SPs). The optimization of fluorescence by tuning the conformation and bulk of side chains, particularly using bulky branched side chains, has been shown to enhance fluorescence intensity and improve photothermal therapy efficacy. The study proposes a feasible molecular guideline for amplifying the NIR-II brightness of SPs for enhanced phototheranostics.

Lanthanide/Cu2-xSe Nanoparticles for Bacteria-Activated NIR-II Fluorescence Imaging of Infection

Leilei Sun et al.

Summary: This study describes a material system that enables specific near-infrared-II (NIR-II) fluorescence imaging of Gram-positive bacteria. The material system is based on the electrostatic binding of copper selenide (Cu2-xSe) and vancomycin-modified nanoparticles, which are weakly fluorescent due to spectral overlap. The presence of Gram-positive bacteria disconnects the bond between the nanoparticles, resulting in a strong fluorescent signal. In vivo studies demonstrate that the material system can specifically detect Gram-positive bacterial infection while showing little fluorescence in the area of Gram-negative bacterial infection.

Summary: Organic dye based NIR-II fluorescent probes offer high signal-to-background ratio and deep-tissue imaging capabilities. A novel class of polymethine dyes, NIRII-RTs, with bright and stable NIR-II emission and large Stokes shifts have been designed. These dyes, particularly NIRII-RT3 and NIRII-RT4, can serve as effective platforms for activatable bioimaging probes. Target-activatable NIRII-RT probes for pH, ATP, and metal-ion detection were synthesized, demonstrating real-time monitoring of drug-induced hepatotoxicity.
